IP6K1 rewires LKB1 signaling to mediate hyperglycemic endothelial senescence.

Diabetes

January 2025

Institute for Developmental and Regenerative Cardiovascular Medicine, Xinhua Hospital, Shanghai Jiao Tong University School of Medicine, Shanghai 200092, China.

Diabetes is a major risk factor for cardiovascular disease, but the molecular mechanisms underlying diabetic vasculopathy have been elusive. Here we report that inositol hexakisphosphate kinase 1 (IP6K1) mediates hyperglycemia-induced endothelial senescence by rewiring the liver kinase B1 (LKB1) signaling from activating the adenosine monophosphate-activated protein kinase (AMPK) pathway to the p53 pathway. We found that hyperglycemia upregulated IP6K1, which disrupts the Hsp/Hsc70 and carboxyl terminus of Hsc70-interacting protein (CHIP)-mediated LKB1 degradation, leading to increased expression levels of LKB1.

In recent years, with the policy reform of new drug review and approval, China has ushered in a surge of innovative drug development. Based on the Pharnexcloud database and the Pharmcube database, we analyzed the innovative drugs approved for marketing and entered clinical trials in China, sorted out the major research and development (R&D) institutions and enterprises, the distribution of innovative drug target types, and the primary therapeutic areas of the approved innovative drugs, and compared with the global innovative drug R&D landscape. Since 2020, China's innovative drug development has shown a rapid growth trend, with intense competition among biopharmaceutical companies, and the emergence of a number of leading biopharmaceutical enterprises.

Introduction: ICAM-1 is an adhesion molecule expressed on the endothelial cells and is involved in regulating leukocyte recruitment to the site of inflammation. Elevated ICAM-1 mRNA expression was found in the serum of mothers with chorioamnionitis. This study aimed to determine the expression of ICAM-1 in the placenta and umbilical cord of pregnancy with chorioamnionitis, and its association with adverse neonatal outcome.

Melatonin stabilizes atherosclerotic plaques: an association that should be clinically exploited.

Front Med (Lausanne)

Atherosclerosis is the underlying factor in the premature death of millions of humans annually. The cause of death is often a result of the rupture of an atherosclerotic plaque followed by the discharge of the associated molecular debris into the vessel lumen which occludes the artery leading to ischemia of downstream tissue and to morbidity or mortality of the individual. This is most serious when it occurs in the heart (heart attack) or brain (stroke).

Background: Recent studies have indicated that oxidative stress is a crucial pathophysiological process in glaucoma. We hypothesized that Posner-Schlossman syndrome (PSS) is a vascular inflammation in the ciliary body due to oxidative stress and endothelial dysfunction. Thus, we investigated serum uric acid (UA) and lipid levels in patients with refractory PSS with the aim of providing basic evidence for the mechanism of PSS.
