In this contribution the synthesis and characterisation of a series of novel mixed ligand iridium(iii) complexes, functionalised with a carboxy ester or phosphonate groups are reported. These groupings are introduced on the 4-position of either the phenyl pyridine or the 2,2'-bipyridyl ligands. A low temperature high yield synthesis for the precursor [Ir(ppy-COOEt)2(μ-Cl)]2 was developed. The photophysical and electrochemical properties of these compounds are also described, together with their behaviour as photosensitisers for the generation of hydrogen from water.

A series of highly substituted tetrahydrofurans (THFs), decorated with modifiable 2-aryl, 3-carboxy and 4-amino substituents, has been prepared for biological evaluation within the European Lead Factory. Diastereoselective reductive amination of pre-functionalised 4-oxofurans, readily prepared from cinnamate esters via oxa-Michael/Dieckmann annulation, provided the requisite THF cores on gram scale with three contiguous stereocentres, including full substitution at C-3. In a second series, a pyrrolidine ring was fused to the same oxofuran scaffold via an intramolecular reductive amination, inverting the configuration at C-4 relative to the other ring substituents.

Urinary 11-nor-Δ(9)-tetrahydrocannabinol-9-carboxylic acid (Carboxy-THC) concentrations, normalised to creatinine output, have been demonstrated to be a useful tool in the interpretation of the results of a series of urine tests for cannabis. These tests, often termed historical data, can be used to identify potential chronic cannabis users who may present occupational health and safety risks within the workplace. Conversely, the data can also be used to support employee claims of previous regular, rather than recent, cannabis use.

Article Synopsis
  • 2-Oxoglutarate (2OG) and iron-dependent oxygenases are important targets for treating human diseases, and this study evaluates two new inhibitors, IOX1 and 4C8HQ, against common inhibitors like NOG and 2,4-PDCA.
  • The findings show that IOX1 effectively inhibits a wide range of 2OG oxygenases, including those related to histone and nucleic acid demethylation, and works in both cytosolic and nuclear environments without needing chemical modifications.
  • Additionally, unique crystallographic studies reveal that IOX1 can cause significant movement of an active site metal in the oxygenases, which is an unusual phenomenon not seen with 4C8HQ.

A validated method for the quantification of Delta(9)-tetrahydrocannabinol (THC) and its main metabolites 11-hydroxy-tetrahydrocannabinol (OH-THC) and 11-nor-9-carboxy-tetrahydrocannabinol (THC-COOH) in serum is presented. The substances were isolated by solid-phase extraction, derivatised by methylation, and analysed by means of GC-MS in the selected ion monitoring mode. Quantitation was achieved by the addition of deuterated analogues as internal standards.
