Analysis of Type II NAD(P)H Dehydrogenases.

Plant mitochondria contain at least four type II NAD(P)H dehydrogenases that link NAD(P)H oxidation to the inner membrane electron transport chain and bypass proton pumping at Complex I, hence ATP synthesis. These activities have been found in mitochondria isolated from all plant species analyzed to date. In this chapter, methods are presented to analyze the expression of genes encoding these dehydrogenases and to detect protein levels in mitochondria isolated from Arabidopsis (Arabidopsis thaliana). In addition, methods and assay conditions are presented to detect the activity of each of these four type II NAD(P)H dehydrogenases in isolated plant mitochondria.

Liraglutide, a human glucagon-like peptide-1 (GLP-1) analogue, is a potential treatment option for diabetes. This study aimed to examine the potential of liraglutide (LIRA) in inhibiting ferroptosis and reducing high glucose-induced renal fibrotic injury in mice, and whether the Fsp1-CoQ10-NAD(P)H signal pathway is a mechanism for this effect.

: A large amount of recent evidence suggests that cellular inability to consume oxygen could play a notable part in promoting sepsis as a consequence of mitochondrial dysfunction and oxidative stress. The latter could, in fact, represent a fundamental stage in the evolution of the "natural history" of sepsis. Following a study previously conducted by the same working group on heart samples, the present research project aims to evaluate, through an immunohistochemical study, the existence and/or extent of oxidative stress in the brains of subjects who died due to sepsis and define, after reviewing the literature, its contribution to the septic process to support the use of medications aimed at correcting redox anomalies in the management of septic patients.

The peptidoglycan biosynthetic pathway involves a series of enzymatic reactions in which UDP-N-acetylglucosamine-enolpyruvate reductase (MurB) plays a crucial role in catalyzing the conversion of UDP-N-acetylglucosamine-enolpyruvate (UNAGEP) to UDP-N-acetylmuramic acid. This reaction relies on NADPH and FAD and, since MurB is not found in eukaryotes, it is an attractive target for the development of antimicrobials. MurB from Brucella ovis, the causative agent of brucellosis in sheep, is characterized here.
