The Major Histocompatibility Complex (MHC) is a cluster of genes with primarily immune-related functions. The MHC class I genes are responsible for self- versus non-self-recognition and viral antigen presentation to T lymphocytes. The chicken MHC class I protein binds its cognate antigen(s) over a repertoire spectrum ranging from promiscuous (generalist) to fastidious (specialist).

Exploring red blood cells as an antigen delivery system to modulate the immune response towards FVIII in hemophilia A.

J Thromb Haemost

November 2024

Department of Molecular Hematology, Sanquin Research and Landsteiner Laboratory, Amsterdam, The Netherlands. Electronic address:

Background: The main complication in hemophilia A treatment is the development of inhibitory antibodies against factor (F)VIII. Immune tolerance induction, the gold standard for eradicating anti-FVIII antibodies, is efficient in only 60% to 80% of cases. This underscores the need for more efficient induction of tolerance in patients with hemophilia A with FVIII inhibitors.

Antigen presentation for central tolerance induction.

Nat Rev Immunol

January 2025

Institute for Immunology, Biomedical Center (BMC), Faculty of Medicine, LMU Munich, Planegg-Martinsried, Germany.

The extent of central T cell tolerance is determined by the diversity of self-antigens that developing thymocytes 'see' on thymic antigen-presenting cells (APCs). Here, focusing on insights from the past decade, we review the functional adaptations of medullary thymic epithelial cells, thymic dendritic cells and thymic B cells for the purpose of tolerance induction. Their distinct cellular characteristics range from unconventional phenomena, such as promiscuous gene expression or mimicry of peripheral cell types, to strategic positioning in distinct microenvironments and divergent propensities to preferentially access endogenous or exogenous antigen pools.

Article Synopsis
  • Rotavirus, a segmented dsRNA virus, has its VP1 gene encoding RNA-dependent RNA polymerase (RdRp), which is targeted for a potential multiepitope vaccine using immunoinformatic methods.
  • The study analyzed 100 nucleotide sequences from various global strains to create a consensus sequence, identifying 27 highly immunogenic and promising T-cell and B-cell epitopes for the vaccine.
  • Results show strong binding interactions and immunogenic responses for the multiepitope vaccine, indicating its potential effectiveness in reducing rotavirus infections globally.
