Objective To investigate the protective effect of curcumin (Cur) against arsenic-induced neuroimmune toxicity and the underlying molecular mechanisms in vivo. Methods Eighty SPF female C57BL/6 mice were randomly assigned to four groups: a control group, an arsenic-treated group, a Cur-treated group and an arsenic+Cur group, with 20 mice in each group. The control group received distilled water; the arsenic-treated group was given 50 mg/L NaAsO in the drinking water; the Cur-treated group was gavaged with 200 mg/kg of curcumin for 45 days; and the arsenic+Cur group received distilled water and was gavaged with 200 mg/kg of curcumin.

Retinoid-related orphan receptor-γ (RORγ) is a nuclear receptor that plays important roles in the development and activation of T helper type-17 (Th17) cells. In this study, we characterized the pharmacological profile of JTE-151 ((4S)-6-[(2-chloro-4-methylphenyl)amino]-4-{4-cyclopropyl-5-[cis-3-(2,2-dimethylpropyl)cyclobutyl]isoxazol-3yl}-6-oxohexanoic acid), which is a novel RORγ antagonist identified by our group. JTE-151 dissociated co-activator peptide from the human RORγ-ligand binding domain (LBD) and recruited co-repressor peptide into human RORγ-LBD, and potently inhibited the transcriptional activity of RORγ of human, mouse and rat.

Distinct RORγt-dependent Th17 immune responses are required for autoimmune pathogenesis and protection against bacterial infection.

T helper (Th)17 cells mediate both protective anti-bacterial immune responses and autoimmune pathogenesis, but the distinct pathways regulating these Th17 responses remain unclear. Retinoid-related orphan receptor γ t (RORγt) is a master transcription factor that governs Th17 cell generation and effector functions. We found that a K256R mutation in RORγt impairs Th17-mediated experimental autoimmune encephalomyelitis (EAE) without affecting the clearance of Citrobacter rodentium.

Article Synopsis
  • Sepsis-induced immune system abnormalities can lead to organ dysfunction, and maintaining the balance between Th17 and Treg cells is crucial for immune health.
  • The study used mouse models and various scientific methods to evaluate the effects of bergapten, a natural compound, on inflammation during sepsis.
  • Results showed that bergapten reduced lung damage and inflammation by restoring the Th17/Treg balance, suggesting its potential as a treatment for sepsis-related conditions.

Article Synopsis
  • Psoriasis is a chronic skin disease linked to an abnormal immune reaction and oxidative stress, which can worsen due to sleep disturbances.
  • The Xiaoyin-Anshen formula (XYAS), a traditional Chinese medicine, shows promise in treating psoriasis, especially for patients suffering from sleep issues, but its exact effects are not fully understood.
  • A study using a mouse model aimed to investigate how XYAS influences melatonin levels, reduces oxidative stress, and mitigates psoriasis-like inflammation through a series of treatments and tests.
