Objective: To investigate the influence of sulfur-fumigated Paeoniae Radix Alba on the contents of main constituents in Guizhi Decoction.
Methods: HPLC was applied to the simultaneous determination and comparative analysis of ten main indicative components in Guizhi Decoction contained sun-dried and sulfur-fumigated Paeoniae Radix Alba. The analysis was performed on a Kromasil C18 column (250 mm x 4.6 mm, 5 μm) with the mobile phase consisting of acetonitrile-0.1% aqueous formic acid. The flow rate was 1.0 mL/min and the column temperature was 30 °C. The detection wavelengths were set at 254 nm and 280 nm, respectively. 20 μL was injected for analysis with gradient elution.
Results: There were obvious differences existed between the concentrations of ten main indicative components in Guizhi Decoction contained sun-dried and sulfur-fumigated Paeoniae Radix Alba.
Conclusion: Sulfur-fumigated Paeoniae Radix Alba can influence the main effective constituents contained in Guizhi Decoction in quantity. The absence in the study of prescriptions affected by sulfur-fumigation is filled up through the investigation of the chemical basis of Guizhi Decoction influenced by sulfur-fumigated Paeoniae Radix Alba, and also strong evidences are offered to the further regulation and supervision of sulfur-fumigation in herbal medicines.

Introduction: Sulfur-fumigation of Paeoniae Radix Alba (PRA) could induce the chemical transformation of its bioactive component paeoniflorin into a sulfur-containing derivative paeoniflorin sulfite, and thus alter the quality, bioactivities, pharmacokinetics, and toxicities of PRA. However, how sulfur-fumigated PRA (S-PRA) affects the quality of PRA-containing complex preparations has not been intensively evaluated.
Objectives: We intend to evaluate the influence of S-PRA on the overall quality of three kinds of Si-Wu-Tang (SWT) formulations, i.

Peony root is an important herbal drug used as an antispasmodic analgesic. To evaluate peony roots with different botanical origins, producing areas, and post-harvest processing, H NMR-based metabolomics analysis was employed. Five types of monoterpenoids, including albiflorin (4), paeoniflorin (6), and sulfonated paeoniflorin (25), and six other compounds, including 1,2,3,4,6-penta-O-galloyl-β-D-glucose (18), benzoic acid (21), gallic acid (22), and sucrose (26) were detected in the extracts of peony root samples.

Since the current identification method for Paeoniae Radix Alba is complex in operation and long time-consuming with high requirements for technicians, the present study employed Heracles NEO ultra-fast gas phase electronic nose(E-nose) technology to identify raw and sulfur-fumigated Paeoniae Radix Alba decoction pieces in order to establish a rapid identification method for sulfur-fumigated Paeoniae Radix Alba. The odors of raw Paeoniae Radix Alba and its sulfur-fumigated products were analyzed by Heracles NEO ultra-fast gas phase E-nose to obtain the odor chromatographic information. The chemometric model was established, and the data were processed by principal component analysis(PCA), discriminant function analysis(DFA), soft independent modeling of class analogy(SIMCA), and partial least squares discriminant analysis(PLS-DA).

In this study, a time segment scanning-based quasi-multiple reaction monitoring (quasi-MRM) mode was proposed to improve the quantitative performance of UPLC-QTOF-MS/MS. To achieve the quasi-MRM mode, a strategy to select the ion pair (precursor and product ions) of each analyte was adopted as follows. First, a stable and abundant ion by quadrupole was set as precursor ion in MS scan mode.

In traditional Chinese medicine practice, crude herbs are often subjected to traditional processing (Paozhi in Chinese) for a special medicinal purpose. Bran-frying is one of processing methods for Paeoniae Radix Alba (PRA). Previous studies found that paeoniflorin and paeoniflorin sulfonate, a principle bioactive compound and its sulfur-fumigation induced characteristic sulfur-containing derivative, could be used together with sulfur dioxide residue as chemical markers for the quality control of sulfur-fumigated PRA crude material.
