An overview of the state-of-the art in HPLC coupled with fluorescence detection is presented. Over the last 20 years, the increasing number of methodological papers on this topic (4082 between 1994 and 2004 and 7725 between 2004 and 2014) is testament to its utility in bioanalytical applications. Compared with conventional UV absorbance detection used in HPLC, fluorescence detection can greatly enhance the sensitivity leading to limits of detection similar to those obtained with mass spectrometry, offering researchers a sensitive, robust and relatively inexpensive instrumental method. This work will focus on the analysis of pharmaceutical compounds in different biological matrices, either naturally fluorescent or derivatized with a fluorescent agent, and some of them chiral. Therapeutic applications, sample preparation and derivatization, sensitivity for each example are described.

Objective To study the impacts of curcumin on the proliferation, migration and cisplatin (DDP) resistance of bladder cancer cells by regulating the liver kinase B1-AMP activated protein kinase-microtubule-associated protein 1 light chain 3 (LKB1-AMPK-LC3) signaling pathway. Methods Human bladder cancer cell line T24 was cultured in vitro, and its DDP resistant T24/DDP cells were induced by cisplatin (DDP). After treating T24 and T24/DDP cells with different concentrations of curcumin, the optimal concentration of curcumin was screened by MTT assay.

Coupling interactions between the alpha (α) subunit of the polymerase III core (α-Pol III core) and the tau (τ) subunit of the clamp loader complex (τ-CLC) are vital for efficient and rapid DNA replication in Escherichia coli (E. coli). Specific and targeted mutations in the C-terminal τ-interaction region of the Pol III α-subunit disrupted efficient coupled rolling circle DNA synthesis in vitro and caused significant genomic defects in CRISPR-Cas9 dnaE edited cell strains.

Determination of hydrogen peroxide (HO) is of great importance in many systems for controlling the quality of products, food safety, and medical diagnostics. In this work, a highly sensitive photoluminescence film sensor was synthesized based on chitosan (CS), polyvinyl alcohol (PVA), and terephthalic acid (TPA), in the presence of copper (II) ions for determination of hydrogen peroxide. TPA was used as a sensitive probe for detection of hydroxyl radicals produced in a photo-Fenton-like process.

pH balance is an important factor in regulating the internal environment of body and maintaining the normal physiological activities, but pH cannot be detected in vivo without damaging the tissue. It is important to develop a pH probe with low toxicity, high sensitivity and targeting of organelles. In this research, a novel carbazole-pyrimidine-based probe PKZP was designed from 2-hydroxyl-3-pinanone which was derived from natural monoterpene α-pinene for detecting both acidic and basic pH in vivo.

Flavonoid glycosides are formed by dehydration condensation of aglycones and sugar molecules. Therefore, discrimination of flavonoid glycosides from their corresponding aglycones is a challenging task because they contain the same aglycone part in their molecular structures. Herein, boric acid-functional Eu(III)-organic framework (BA-Eu-MOF) was applied to discriminate flavonoid glycosides including baicalin (Bai), wogonoside (Wog), rutin (Rut), puerarin (Pue), quercitrin (Que) and astragalin (Ast) from their corresponding aglycones for the first time.
