Published: April 2015

New simple sequence repeat (SSR) markers were developed in the Japanese larch (Larix kaempferi) using unigene sequences for further genetic diversity studies and the genetic improvement of breeding programs. One thousand two handred and thirty five (1235) primer pairs were tested and 165 successfully identified in L. kaempferi. Out of the amplified candidate markers, 145 (90.6%) exhibited polymorphism among 24 individuals of L. kaempferi, with the number of alleles per locus (Na), observed heterozygosity (Ho), expected heterozygosity (He) and polymorphic information content (PIC) averaging at 4.510, 0.487, 0.518 and 0.459, respectively. Cross-species amplification of randomly selection of 30 genic-SSRs among the 145 polymorphic ones showed that 80.0% of the SSRs could be amplified in Larix olgensis, 86.7% could be amplified in Larix principi-rupprechtii and 83.0% could be amplified in Larix gmelinii. High rates of cross-species amplification were observed. The genic-SSRs developed herein would be a valuable resource for genetic analysis of Larix kaempferi and related species, and also have the potential to facilitate the genetic improvement and breeding of larch.

CRISPR/Cas9 ribonucleoprotein mediated DNA-free genome editing in larch.

Here, a DNA-free genetic editing approach is presented for larch by delivering ribonucleoprotein complexes (RNPs) of CRISPR/Cas9 through particle bombardment. The detailed procedure encompasses creating a transgenic system particle bombardment for the transformation of embryogenic callus, validating the functionality of RNPs, optimizing coating and delivery techniques, enhancing somatic embryo maturation, regenerating plantlets, and precisely identifying mutants. The optimal particle bombardment parameters were determined at 1,100 psi and a distance of 9 cm and the editing efficiency of the targets was verified .

Somatic embryogenesis is valuable for clonal propagation and genetic improvement, and it also serves as an ideal system for studying plant development mechanisms. In , microRNA171 and its target gene (), which has two alternative splicing variants, can regulate somatic embryogenesis; however, the underlying molecular mechanism is still unknown. In this study, we overexpressed these two variants in and and then used the RNA-Seq method to screen genes from and , whose expression patterns are related to those of variants.

As an important branch characteristic factor, the quantity of branches could influence crown structure, tree growth, and wood quality. Taking plantation in Dagujia Forest Farm, Qingyuan County, Liao-ning Province as the research object, we developed a mixed effect prediction model of the first-order branches quantity of including sprouting branches based on the negative binomial distribution model, and a mixed effect prediction model of the first-order branches density of including sprouting branches based on the negative exponential model. The results showed that the mixed effect model considering sample level as the random effect effectively decreased the heteroscedasticity and autocorrelation.

Article Synopsis
  • Promoters are essential for creating new plant varieties through transgenic technology, but the inconsistent expression of target genes in Japanese larch genetic transformation is a challenge.
  • This study focused on analyzing transcriptome data to find highly expressed genes, cloning their promoters, and constructing expression vectors with a reporter gene for testing.
  • After introducing these vectors into embryogenic callus through genetic transformation, the study identified four active promoters, with one showing significantly higher activity, suggesting it could be ideal for larch genetic transformation.
