The incorporation of the 4-phenylpiperidine pharmacophore found in morphine into 4-anilidopiperidines related to fentanyl (1) led to a novel class of potent opioid analgesic and anesthetic agents with a favorable pharmacological profile. The synthesis, analgesic activity, and anesthetic properties of a series of 4-phenyl-4-anilidopiperidines (13-29) are discussed. Isosteric replacement of the phenyl by various heteroaryl substituents extended the series to include 4-heteroaryl-4-anilidopiperidines (30-53). Within this group, 1-[2-(1H-pyrazol-1-yl)ethyl]-4-(4-methylthiazol-2-yl)-4-(N- phenylpropionamido)piperidine (48), exhibited high analgesic potency, short duration of action, rapid recovery of motor coordination following anesthetic doses, and greater cardiovascular and respiratory safety during anesthesia as compared with opioids fentanyl (1) and alfentanil (2) currently in clinical use. Such analgesics could be of great utility to clinicians in the expanding outpatient surgical arena and for patient-controlled analgesia and computer assisted continuous infusion pain control techniques.

Objectives: The aim of this study was to examine the marginal seal of novel bioactive restorative materials and the material-related properties associated with bacterial microleakage.
Methods: Class II cavities prepared into human extracted teeth were restored with: Venus Diamond (VD) + selective enamel etching (SEE)/self-etching universal adhesive (SEA), ACTIVA BioACTIVE RESTORATIVE (AB) + SEE/SEA, Cention Forte (CF) + Cention Primer, Ketac Universal Aplicap (KU), EQUIA Forte HT (EF) and Surefil One (SO) and exposed to a cariogenic multi-species bacterial suspension for 7 days. Bacterial microleakage was visualized with a modified gram staining protocol and bacterial penetration depths were microscopically determined after sectioning the teeth.

Mechanisms for absorption improvement of drugs with low water-solubility by self-microemulsifying drug delivery system (SMEDDS) are still controversial except for solubility improvement. We attempted to clarify the mechanisms by utilizing model drugs classified as biopharmaceutics classification system class II. Porphysomes are a class of liposome-like nanoparticles that have demonstrated efficacy in photothermal therapy (PTT) and photodynamic therapy (PDT) against cancer. These nanoparticles results from the self-assembly of amphiphilic phospholipid-porphyrin (PL-Por) conjugates. Despite their potential, porphysomes exhibit a high photothermal effect and a weak photodynamic activity as long as they remain intact within the body.
