The action potential (AP) in most nerve and muscle preparations depends upon nanomolar concentrations of the neurotoxins saxitoxin (STX) and tetrodotoxin (TTX). In some excitable tissues lacking mature innervation, a toxin-resistant AP has been described by electrophysiological results. However, multiple attempts to detect corresponding toxin-resistant Na channels with radiolabelled STX and TTX have been unsuccessful. We report here the detection of Na channels with low-affinity binding of STX and TTX, accounting for 50-60% of the Na channels in rat hindlimb muscle 4-5 days after denervation.

Initial proof-of-concept development of competitive lateral flow (LF) test strips involving red quantum dots (Qdots) is demonstrated with visible confirmation of the competitive displacement of Qdot-protein-saxitoxin (Stx) and -tetrodotoxin (Ttx) conjugates over 15 min. The measured limits of detection (LODs) for the best versions of the assays were in the 1-2 µg range when a bovine serum albumin (BSA)-Stx or -Ttx conjugate was used as the analyte for safety reasons and NIH ImageJ image analysis was applied. However, when one assumes that only one of the primary amines in BSA labeled with Stx or Ttx is needed as the "epitope" for capture and calculates the weight of the toxin adducts on an equal mole basis to that of 1 µg of toxin-BSA conjugate, the LODs are as low as 4.

Aptamers are single-stranded oligonucleotides that fold into defined architectures for specific target binding. In this study, aptamers are selected that specifically bind to small-molecule neurotoxins and encapsulate them into cell membrane-coated nanoparticles (referred to as 'cellular nanoparticles' or 'CNPs') for effective neutralization of neurotoxins. Saxitoxin (STX) and tetrodotoxin (TTX) are widely distributed and extremely harmful marine toxins, it is certainly worth to spend effort to develop facile methods to detect them in sea food for human safety. In this work, two nano-sensors were developed by combining with two zirconium fluorescence Nanoscale metal-organic frameworks (NMOFs) with two emissions and TAMRA-labelled aptamers for STX and TTX sensing, respectively. The recognition of STX and TTX by these nano-sensors could change the structure of aptamer, which caused the blue or green emissions from NMOFs (energy donor) decreased while red emission from TAMRA-labelled aptamers (energy acceptor) increased owing to fluorescence resonance energy transfer (FRET) effect.
