It is found that after administration of 3'-methyl-4-dimethylaminoazobenzene (3'-Me-DAB,) which was hepatocarcinogenic to rats, in suckling mice, the number of neoplastic lesions in the liver of mice was 3 times higher than after analogous administration of equimolar dose of ortho-aminoazotoluene (OAT)). However, in the Ames test (TA-98 strain of Salmonella typhimurium) with activation by hepatic enzymes (S-9 fraction) of both intact and Aroclor-1254-induced mice and rats OAT contributed by an order of magnitude to revertant colonies compared to 3'-Me-DAB. In vivo inhibition of sulfotransferase activity, the enzyme which catalyzes the final stage of the mutagenic activation of aminoazo dyes, had no effect on carcinogenicity of 3'-Me-DAB but more than 4 times elevated that of OAT. It was concluded that the mechanism of carcinogenic action of aminoazo dyes studied is not genotoxic and that the carcinogenic potential of OAT is lost in the process of mutagenic activation.

In the present study, the ability of duckweed (Lemna minor L.) in the decolorization of Acid Bordeaux B (ABB), as an aminoazo benzene dye, from polluted waters was evaluated. It was found that the rise of temperature and enhancement of the plant initial weight led to increasing the dye removal efficiency, but raising the initial dye concentration and pH reduced it.

4-Dimethylaminoazobenzene (DAB, methyl yellow, or butter yellow), a human carcinogen, has been banned for use in foods since 1988. In 2014, DAB adulteration in Tofu occurred in Taiwan. We hypothesize that DAB can form [DAB•SBP]adduct adduct with soybean protein (SBP) which could damage Gastro-Duodenal-Hepatic axis.

It was found that in the Ames test they have mutagenic activity, however, this activity does not correlate quantitatively with rat sensitivity to their hepatocarcinogenic action. Thus, the most active carcinogen 3'-methyl-4-dimethylaminoazobenzene causes mutations almost 2.

A new hypothesis is discussed, which describes the initiation of the carcinogenesis through polycyclic aromatic hydrocarbons (PAHs) and aminoazo dyes (AZOs) as a two-step process: the oncogenic proteins of the ras or ras-like on-cogenes activated by mutation ("initiation A ") co-operate with the complexes in the plasma membrane formed during the "initiation B " stage from the parent compounds of the PAHs or AZOs with cholesterol and apolipoprotein A-I. The final result of this co-operation, or the "complete initiation", is an irreversibly modified membrane architecture with negative consequences for growth control.
