The fast response to stimuli and subsequent activation of the nuclear factor of activated T cells (NFAT) signaling pathway play an essential role in human T cell functions. MicroRNAs (miRNAs) are increasingly implicated in regulation of numerous biological and pathological processes. In this study we demonstrate a novel function of miRNA-9 (miR-9) in regulation of the NFAT signaling pathway. Upon PMA-ionomycin stimulation, miR-9 was markedly increased, consistent with NFAT activation. Overexpression of miR-9 significantly enhanced NFAT activity and accelerated NFAT dephosphorylation and its nuclear translocation in response to PMA-ionomycin. Karyopherin-β1 (KPNB1, a nucleocytoplasmic transporter) and dual-specificity tyrosine phosphorylation-regulated kinase 1B (DYRK1B) were identified as direct targets of miR-9. Functionally, miR-9 promoted IL-2 production in stimulated human lymphocyte Jurkat T cells. Collectively, our data reveal a novel role for miR-9 in regulation of the NFAT pathway by targeting KPNB1 and DYRK1B.

Thermoelectric (TE) performance in materials is often constrained by the strong coupling between carrier and phonon transport, necessitating trade-offs between electrical and thermal properties that limit improvements in the figure of merit (). Herein, a novel strategy is proposed to achieve simultaneous energy filtering and enhanced phonon scattering, effectively optimizing the TE properties of CoSb-based skutterudites. By introducing CuTe nanoprecipitates into the YbCoSb matrix, interfacial barriers are formed, which selectively filter low-energy charge carriers, significantly improving the Seebeck coefficient while maintaining high carrier mobility.

Background: The two-stage surgical technique of associated liver partition and portal vein ligation for staged hepatectomy (ALPPS) enables extensive liver resection and promotes future liver remnant regeneration (FLR), in part by inhibiting the Hippo signalling pathway. Its main effector, Yes-associated protein (YAP), has low intrinsic transcriptional activity and requires the transcription enhanced associated domain factor (TEAD) family members as cofactors for target gene transcription. Purpose: To investigate the prognostic value of post-chemoradiotherapy 2-[F]FDG PET/CT in locally advanced nasopharyngeal carcinoma (LANPC) and develop an accurate prognostic model based on the 2-[F]FDG PET/CT results.
Methods: 900 LANPC patients who underwent pretreatment and post-chemoradiotherapy 2-[F]FDG PET/CT from May 2014 to August 2022 were included in the study. We divided the patients into two distinct cohorts for the purpose of our study: a training cohort comprising 506 individuals, included from May 2008 to April 2020, and a validation cohort consisting of 394 individuals, included from May 2020 to August 2022.
