Presence of virulent Newcastle disease virus in vaccinated chickens in farms in Pakistan.

Published: May 2015

One year after a virulent Newcastle disease virus (vNDV) outbreak in Pakistan, the causative strain was present in vaccinated chickens of multiple farms despite the existence of high-average NDV-specific antibody titers (>4.75 log2). The data suggest a possible role of vaccinated birds as reservoirs of vNDV.

Article Synopsis
  • Newcastle disease virus (NDV) serves as a valuable model for studying viral mechanisms and is being utilized as a vector for vaccine development and cancer therapy.
  • A reverse genetic system was created for the NDV Mukteswar strain, allowing for the insertion of the enhanced green fluorescent protein (EGFP) gene between the P and M genes to form a stable transcription unit that expresses exogenous genes over multiple generations.
  • Through this system, a recombinant virus that co-expresses EGFP and cherry fluorescent protein (CFP) was developed, demonstrating efficient simultaneous expression of both proteins, though with slight differences in expression levels, and similar growth rates with mild virulence attenuation compared to the original strains.

Newcastle disease (ND) is among the most common and deadliest poultry diseases worldwide. Thermostable Newcastle disease virus (NDV) vaccines have been widely used to protect village chickens against ND due to their decreased dependence on cold chains for transport and storage. The NDV4 Heat-Resistant (NDV4HR) vaccine is an apathogenic, heat-resistant, live vaccine that can induce immunity in chickens.

Pigeon Newcastle disease (ND) is the most common viral infectious disease in the pigeon industry, caused by pigeon paramyxovirus type 1 (PPMV-1), a variant of chicken-origin Newcastle disease virus (NDV). Previous studies have identified significant amino acid differences between PPMV-1 and chicken-origin NDV at positions 347 and 349 in the hemagglutinin-neuraminidase (HN) protein, with PPMV-1 predominantly exhibiting glycine (G) at position 347 and glutamic acid (E) at position 349, while most chicken-origin NDVs show E at position 347 and aspartic acid (D) at position 349. However, the impact of these amino acid substitutions remains unclear.

Stress contingent changes in Hog1 pathway architecture and regulation in Candida albicans.

The Hog1 stress-activated protein kinase (SAPK) is a key mediator of stress resistance and virulence in Candida albicans. Hog1 activation via phosphorylation of the canonical TGY motif is mediated by the Pbs2 MAPKK, which itself is activated by the Ssk2 MAPKKK. Although this three-tiered SAPK signalling module is well characterised, it is unclear how Hog1 activation is regulated in response to different stresses.

Bi-allelic variants in DAP3 result in reduced assembly of the mitoribosomal small subunit with altered apoptosis and a Perrault-syndrome-spectrum phenotype.

The mitochondrial ribosome (mitoribosome) synthesizes 13 protein subunits of the oxidative phosphorylation system encoded by the mitochondrial genome. The mitoribosome is composed of 12S rRNA, 16S rRNA, and 82 mitoribosomal proteins encoded by nuclear genes. To date, variants in 12 genes encoding mitoribosomal proteins are associated with rare monogenic disorders and frequently show combined oxidative phosphorylation deficiency.
