A strategy for the electrochemical detection of DNA based on catalytic hairpin assembly combined with nanocatalyst label-based redox cycling reaction signal amplification. A superior detection limit of 0.3 aM toward DNA can be achieved.

A novel detection technique is introduced that offers sensitive and reliable ochratoxin A (OTA) detection. The method leverages the etching of gold nanorods (AuNRs) stabilized by hexadecyl trimethyl ammonium bromide (CTAB) using the oxidized form of 3,3',5,5'-tetramethyl benzidine sulfate (TMB), creating a susceptible multicolor visual detection system for OTA. The visual detection is enabled by Mg-assisted DNAzyme catalysis combined with the catalytic hairpin assembly (CHA) signal amplification strategy.

PH-20 is a specific type of hyaluronidase that plays a critical role in the fertilization process by facilitating the initial binding of sperm to the glycoprotein layer surrounding the oocyte and subsequently breaking down hyaluronic acid polymers in the cumulus cell layer. PH-20 contains an epidermal growth factor (EGF)-like domain, which may be involved in the recognition of the glycoprotein layer in addition to the catalytic domain. Herein, we report the structure of human PH-20 determined by cryogenic electron microscopy.

Currently, metal-organic frameworks (MOFs) with tunability and covalent organic frameworks (COFs) with high stability are promising nanomaterials for electrochemiluminescence (ECL), while Ru-based metal covalent organic frameworks (Ru-MCOFs) have rarely been reported. Herein, an ECL immunosensor based on a strong ECL-emitting Ru-MCOF was proposed for the sensitive detection of the cardiac troponin-I (cTnI). Imine-linked Ru-MCOF was prepared as an ECL emitter via solvothermal method using tris(4,4' -diamino-2,2' -bipyridine)ruthenium(II) (Ru(dbpy)32+) as a precursor.

Cytochromes P450 (CYP) form one of the largest enzyme superfamilies on Earth, with similar structural fold but biological functions varying from synthesis of physiologically essential compounds to metabolism of myriad xenobiotics. Here we determined the crystal structures of and human sterol 14α-demethylases (CYP51s). Both structures reveal elements that imply elevated conformational flexibility, uncovering molecular basis for faster catalytic rates, lower substrate selectivity, and resistance to inhibition.
