Receptors of the thyroid: the thyrotropin receptor is only the first violinist of a symphony orchestra.

A basic reason for undertaking these studies was to further our knowledge of the structure and function of the TSH receptor as well as its interaction with other receptors on thyroid cells. The multiplicity of observations suggests the approach is bearing fruit, does not provide a simple answer, and can have pitfalls. We hope they may also contribute to understanding the structure and function of autoantigens in Graves' disease and glycoprotein hormone receptors in general. The authors are grateful to their collaborators in the National Dental Institute, particularly Drs. Bellur Prabhakar, Edward Oates, and Abner Notkins, in the National Cancer Institute, Drs. W. O. McBride and M. Lerman for their contributions to the cloning studies.

Clinical Relevance and Drug Modulation of PPAR Signaling Pathway in Triple-Negative Breast Cancer: A Comprehensive Analysis.

PPAR Res

December 2024

Department of Laboratory Medicine, The Sixth School of Clinical Medicine, The Affiliated Qingyuan Hospital (Qingyuan People's Hospital), Guangzhou Medical University, Qingyuan, China.

Triple-negative breast cancer (TNBC) is highly heterogeneous and poses a significant medical challenge due to limited treatment options and poor outcomes. Peroxisome proliferator-activated receptors (PPARs) play a crucial role in regulating metabolism and cell fate. While the association between PPAR signal and human cancers has been a topic of concern, its specific relationship with TNBC remains unclear.

This study aimed to investigate the effect of Fuzheng Yiai Decoction (FZYA) on epidermal growth factor receptor-tyrosine kinase inhibitor (EGFR-TKI) drug resistance in lung adenosquamous carcinoma (ASC). The expression of thyroid transcription factor 1 (TTF1) and p63 in tumor cells was observed by immunofluorescence staining. Meanwhile, 25 nude mice successfully inoculated with the human lung ASC cell line NCI-H596 were randomly divided into five groups, namely, the model, gefitinib, low-, medium-, and high-dose FZYA with gefitinib groups.

Resmetirom is a thyroid hormone receptor agonist that has been recently approved by the FDA for the management of metabolic dysfunction-associated steatohepatitis (MASH). MASH is a severe form of metabolic dysfunction-associated fatty liver disease (MASLD), which is marked by hepatic inflammation and potential progression to cirrhosis and liver cancer. This review analyzes and demonstrates the efficacy of resmetirom in reducing intra-hepatic lipids, improving liver histology, and improving metabolic parameters.

CDK4/6 inhibitors in HR-positive breast cancer immunotherapy.

Bioorg Chem

December 2024

Department of General Surgery, Affiliated Hospital of Zunyi Medical University, Zunyi, China; Department of Thyroid and Breast Surgery, Affiliated Hospital of Zunyi Medical University, Zunyi, China. Electronic address:

Breast cancer is the most prevalent malignant tumour among women. Approximately 70 % of patients are hormone receptor (HR)-positive and undergo endocrine therapy as the main form of treatment; however, the efficacy of this type of therapy is limited by some factors, such as drug resistance and complex tumour microenvironments. Using network pharmacology and molecular docking, this study examined how CDK4/6 inhibitors enhance the effects of immunotherapy for HR-positive breast cancer, focusing on their effects on the tumour microenvironment (TME) and immune cell activity.
