Activation-induced cytidine deaminase (AID) initiates class switch recombination (CSR) and somatic hypermutation (SHM) by deaminating cytosine residues in immunoglobulin genes (Igh, Igκ, and Igλ). At a lower frequency, AID also causes collateral DNA damage at non-Ig loci, including genes that are rearranged or mutated in B-cell lymphoma. Precisely how AID is recruited to these off-target sites is not entirely understood. To gain further insight into how AID selects its targets, we compared AID-mediated translocations in two different cell types, B cells and mouse embryonic fibroblasts (MEFs). AID targets a distinct set of hotspots in the two cell types. In both cases, hotspots are concentrated in highly transcribed but stalled genes. However, transcription alone is insufficient to recruit AID activity. Comparison of genes similarly transcribed in B cells and MEFs but targeted in only one of the two cell types reveals a common set of epigenetic features associated with AID recruitment in both cells. AID target genes are enriched in chromatin modifications associated with active enhancers (such as H3K27Ac) and marks of active transcription (such as H3K36me3) in both fibroblasts and B cells, indicating that these features are universal mediators of AID recruitment.

Catch bonds are molecular bonds that last longer under force than slip bonds, which become shorter-lived under force. Although catch bonds were initially discovered in studies of leukocyte and bacterial adhesions two decades ago, they have since been found in many other contexts, including platelet binding to blood vessel walls during clotting, structural support within the cell and between cells, force transmission in the cell's machineries for motility and mechanotransduction, viral infection of host cells, and immunoreceptor mechanosensing. Catch bonds are strengthened by increasing force, which induces structural changes in one or both interacting molecules either locally or allosterically to enable additional contacts at their binding interface, thus lengthening bond lifetimes.

This study investigates innovative hybrid perovskite solar cells using rubidium-germanium-iodide (RbGeI) as the substrate, incorporating various hole transport layers like CuO, CuO, and SnSe, and wide-bandgap chalcogenide electron transport layers (ETLs) like IGZO, WS, InS, and ZnSe. After selecting IGZO as the optimal ETL, its depth was optimized using the SCAPS-1D simulator to evaluate device performance. Three device configurations were examined: device-I (Al/FTO/IGZO/RbGeI/CuO/Ni), device-II (Al/FTO/IGZO/RbGeI/CuO/Ni), and device-III (Al/FTO/IGZO/RbGeI/SnSe/Ni), with a detailed analysis of the doping concentration, thickness of the layer, density of defect, operational temperature, and interface defects.
