Isoenzyme c of horseradish peroxidase (HRP-C) is widely used in enzyme immunoassay combined with chemiluminescence (CL) detection. For this application, HRP-C activity measurement is usually based on luminol oxidation in the presence of hydrogen peroxide (H2O2). However, this catalysis reaction was enhancer dependent. In this study, we demonstrated that Jatropha curcas peroxidase (JcGP1) showed high efficiency in catalyzing luminol oxidation in the presence of H2O2. Compared with HRP-C, the JcGP1-induced reaction was enhancer independent, which made the enzyme-linked immunosorbent assay (ELISA) simpler. In addition, the JcGP1 catalyzed reaction showed a long-term stable CL signal. We optimized the conditions for JcGP1 catalysis and determined the favorable conditions as follows: 50 mM Tris buffer (pH 8.2) containing 10 mM H2 O2, 14 mM luminol and 0.75 M NaCl. The optimum catalysis temperature was 30°C. The detection limit of JcGP1 under optimum condition was 0.2 pM. Long-term stable CL signal combined with enhancer-independent property indicated that JcGP1 might be a valuable candidate peroxidase for clinical diagnosis and enzyme immunoassay with CL detection.

Conventional dual-signal electrochemiluminescence (ECL) sensors feature high sensitivity and reliability, but the involvement of coreactants inevitably results in a complex configuration and shows reproducibility risk. Here, we propose an exogenous coreactant-free dual-signal platform, comprising luminol (anodic luminophore), CdSe quantum dots (cathodic luminophore), and CoO/TiC electrocatalyst (coreaction promoter). At different redox potentials, CoO/TiC induces water oxidation and oxygen reduction to produce OH and O radicals, which subsequently drive cathodic and anodic ECL emission, respectively.

The sensitive detection of inflammatory biomarkers in gingival crevicular fluid (GCF) is highly desirable for the evaluation of periodontal disease. Luminol-based electrochemiluminescence (ECL) immunosensors offer a promising approach for the fast and convenient detection of biomarkers. However, luminol's low ECL efficiency under neutral conditions remains a challenge.

Background: Drug enantiomers often display distinguishable or even opposite pharmacological and toxicologic activities. Therefore it is of great necessity to discriminate enantiomers for guaranteeing safetyness and effectiveness of chiral drugs. Facile chiral discrimination has long been a noticeable challenge because of the minimal differences in physicochemical properties of enantiomers.

The advancement of lanthanide fingerprint sensors characterized by targeted emission responses and low self-fluorescence interference for the detection of biothiols is of considerable importance for the early diagnosis and treatment of cancer. Herein, the lanthanide "personality function tailoring" HOF composite sensor array is designed for the specific discrimination of biothiols (GSH, Cys, and Hcy) based on the activation of various luminescent molecules, such as r-AuNCs/luminol via HOF surface proximity. Lumi-HOF@Ce serves as a versatile platform for catalyzing the oxidation of -phenylenediamine (OPD) to generate yellow fluorescent oligomers, accompanied by the fluorescence attenuation of luminol.

Metastasis, the leading cause of mortality in cancer patients, presents challenges for conventional photodynamic therapy (PDT) due to its reliance on localized light and oxygen application to tumors. To overcome these limitations, a self-sustained organelle-mimicking nanoreactor is developed here with programmable DNA switches that enables bio-chem-photocatalytic cascade-driven starvation-photodynamic synergistic therapy against tumor metastasis. Emulating the compartmentalization and positional assembly strategies found in living cells, this nano-organelle reactor allows quantitative co-compartmentalization of multiple functional modules for the designed self-illuminating chemiexcited PDT system.
