In their native form peptides are often found as oligomeric complexes, meaning they consist of more than one peptide chain. Coiled coils and helical bundles are common examples of such complexes. Their oligomeric state needs to be known precisely as this tremendously influences their biochemical and biophysical properties. The extensive analysis of circular dichroism spectroscopic data is commonly used to investigate the thermodynamics of binding and folding of these complexes. Here we present FitDis! an easy-to-use programme, which fits the most common two-state unfolding transition to the measured thermal unfolding curves of any oligomer of any stoichiometry. We demonstrate, with simulated and real examples, that the comparison of different stoichiometric models fitted to the same dataset reveals the oligomeric states of these complexes along with detailed thermodynamic information. This method will significantly ease the analysis of and increase the amount of information gained from, the thermal unfolding curves of peptide complexes.

αβT cells protect vertebrates against many diseases, optimizing surveillance using mechanical force to distinguish between pathophysiologic cellular alterations and normal self-constituents. The multi-subunit αβT-cell receptor (TCR) operates outside of thermal equilibrium, harvesting energy via physical forces generated by T-cell motility and actin-myosin machinery. When a peptide-bound major histocompatibility complex molecule (pMHC) on an antigen presenting cell is ligated, the αβTCR on the T cell leverages force to form a catch bond, prolonging bond lifetime, and enhancing antigen discrimination.

Virulence of many gram-negative bacteria relies upon delivery of type three effectors into host cells. To pass through the conduit of secretion machinery the effectors need to acquire an extended conformation, and in many bacterial species specific chaperones assist in this process. In plant pathogenic bacterium Pseudomonas syringae, secretion of only few effectors requires the function of chaperones.

In this study, a novel acid-induced heat-set soy protein hydrolysate (SPH) gel was successfully developed. The effects of protein (7 and 8 wt%) and glucono-δ-lactone (GDL, 4, 6, 8, and 10 wt%) concentrations on its aggregation and gelation behaviors were investigated by evaluating the structural, rheological, textural, and physical properties of the SPH gel. The structural properties revealed that GDL promoted the formation of SPH aggregates and gels, primarily via disulfide bonds and hydrophobic interactions, which were closely related to the unfolding of the protein structure, exposed hydrophobic groups, decreased protein solubility, and increased particle size and turbidity during the heating process.

Here we explore the interplay between physical and chemical perturbants to unravel links among native folding, amorphous and ordered aggregation scenarios in IFABP (rat intestinal fatty acid binding protein). This small beta-barrel protein undergoes amyloid-like aggregation above 15 % v/v trifluoroethanol. Our aim was to address the influence of sub-aggregating TFE concentrations on the unfolding transitions of IFABP.
