In this study, cross-linked chitosan (CCS)/bentonite (BT) composite was prepared by the intercalation of chitosan in bentonite and the cross-linking reaction between chitosan and glutaraldehyde. CCS/BT composite was characterized by using Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), X-ray diffraction (XRD) and thermal gravimetric analyses (TGA). Their adsorption characteristics were assessed by using an azo dye (Amido Black 10B) as a model adsorbate. The adsorption of Amido Black 10B onto the CCS/BT composite was found to be optimal at pH 2. The adsorption isotherm was well described by the Langmuir model and the maximum adsorption capacity was 323.6 mg/g at 293K and pH 2. Amido Black 10B adsorption kinetics followed a pseudo-second-order kinetic model. The calculated thermodynamic parameters showed that the adsorption of Amido Black 10B by CCS/BT composite was spontaneous and endothermic in nature.

Fingermark impressions in blood are commonly encountered at violent crime scenes and represent a critical trace that can link an individual to the scene. A range of techniques are available for detecting and enhancing bloody impressions; however, many chemical methods involve using hazardous solvents or require alternative light sources to visualise fluorescence. The use of protein stains to enhance fingermark ridge detail in blood is a common technique used by forensic practitioners around the world. Amido Black is one of the most favoured protein stains due to its strong staining ability. The most common formulation of Amido Black is methanol based, with an ability to simultaneously fix and stain the blood impression, however methanol is toxic and can disrupt some surfaces, potentially compromising fingermark detail.

Metaproteomics represents a promising and fast method to analyze the taxonomic and functional composition of biogas plant microbiomes. However, metaproteomics sample preparation and bioinformatics analysis is still challenging due to the sample complexity and contaminants. In this chapter, a tailored workflow including sampling, phenol extraction in a ball mill, amido black protein quantification, FASP digestion, LC-MS/MS measurement as well as bioinformatics and biostatistical data evaluation are here described for the metaproteomics advancements applied to biogas plant samples.
