Overexpression of human α-synuclein gene in Drosophila can reduce lifespan, and we have performed lifespan assay for A30P and A53Tα-synuclein transgenic and control (elav-GAL4, UAS-A30P, UAS-A53T) flies. Our results showed reduced lifespan of transgenic flies compared to controls. We have also investigated behavioral responses, levels of reactive oxygen species (ROS) and lipid peroxidation (LPO) and activities of catalase (CAT) and superoxide dismutase (SOD) in a combined genetic-toxin model (Ethanol-A30P or A53Tα-synuclein models) and controls. Our results showed that sedation time (ST50) of A30P or A53Tα-synuclein PD model flies was significantly lower while recovery time (RC50) of them was remarkably higher compared to control flies. The levels of oxidative markers (ROS and LPO) were significantly higher and the activities of CAT and SOD were lower in transgenic flies that underwent ethanol exposure compared to control. Based on our earlier studies on antioxidant properties of isolated and characterized molecules from Decalepis hamiltonii (Dh) root extract, its protective effect in this combined toxicity model has been investigated. Surprisingly, Dh treatment increased ST50 and decreased RC50 values of transgenic flies. Moreover, we showed that Dh pre-treatment could decrease the levels of ROS and LPO and increase the activities of CAT and SOD in the ethanol-α-synuclein model. This is the first report on protective effects of natural antioxidants in A30P or A53Tα-synuclein PD model flies against oxidative stress induced by ethanol.

Background/objectives: α-Synuclein (α-syn) protein is a major pathological agent of familial Parkinson's disease (PD), and its levels and aggregations determine neurotoxicity in PD pathogenesis. Although the pathophysiological functions of α-syn have been extensively studied, its biological functions remain elusive, and there are reports of wild-type (WT) α-syn and two missense mutations of α-syn (A30P and A53T) inducing protective neuritogenesis through neurite outgrowth. However, the function of another α-syn mutation, E46K, has not been fully elucidated.

Pathological significance of interaction of Synphilin-1 with mutated alpha-synuclein is well known to have serious consequences in causing the formation of inclusion bodies that are linked to Parkinson's disease (PD). Information extracted so far pointed out that specific mutations, A53T, A30P, and E46K, in alpha-synuclein promote such interactions. However, a detailed structural study of this interaction is pending due to the unavailability of the complete structures of the large protein Synphilin-1 of chain length 919 residues and the mutated alpha-synuclein having all the reported specific mutations so far.

Accumulating evidence demonstrates that alpha-synuclein (α-syn) pathology associated with Parkinson's disease (PD) is not limited to the brain, as it also appears in a select number of peripheral tissues including the liver. In this study, we identified a number of PD-associated α-syn post-translational modifications in the livers of (Thy-1)-h[A30P] mice, a mouse model of familial PD expressing human α-syn harboring the A30P mutation driven by a neuron-specific promoter. , we also demonstrate that human hepatocytes induce post-translational modifications following α-syn fibrillar (PFF) treatment.
