Glioblastoma multiforme (GBM) is a malignant and incurable glial brain tumour. The current best treatment for GBM includes maximal safe surgical resection followed by concomitant radiotherapy and adjuvant temozolomide. Despite this, median survival is still only 14-16 months. Mechanisms that lead to chemo- and radio-resistance underpin treatment failure. Insights into the DNA repair mechanisms that permit resistance to chemoradiotherapy in GBM may help improve patient responses to currently available therapies.

The soft-rock roadways in kilometer-deep coal mines are often damaged by large deformation and have to be periodically expanded and repaired, which seriously restricts the safe and efficient production of coal mines. A typical soft-rock roadway in a kilometer-deep coal mine is selected as the engineering, and the main reasons for roadway deformation are analyzed, and the ground stress and mechanical characteristics are obtained. The Flac numerical model, which can accurately reflect the deformation characteristics of surrounding rock in kilometer-deep soft-rock roadway, has been constructed, and the evolution laws of stress field and its damage mechanism have been analyzed with the vertical stress, vertical displacement and plastic zone.

The regressed arms of reversed replication forks exhibit structural similarities to one-ended double-stranded breaks and need to be protected against uncontrolled nucleolytic degradation. Here, we identify MSANTD4 (Myb/SANT-like DNA-binding domain-containing protein 4), a functionally uncharacterized protein that uniquely counters the replication protein A (RPA)-Bloom (BLM)/Werner syndrome helicase (WRN)-DNA replication helicase/nuclease 2 (DNA2) complex to safeguard reversed replication forks from detrimental degradation, independently of the breast cancer susceptibility proteins (BRCA1/2)-DNA repair protein RAD51 pathway. MSANTD4 specifically interacts with the junctions between single-stranded DNA (ssDNA) and double-stranded DNA (dsDNA) in DNA substrates harboring a 3' overhang, which resemble the structural features of regressed arms processed by WRN-DNA2.
