Stimulation of Escherichia coli with acetate elevates the acetylation level of the chemotaxis response regulator CheY. This elevation, in an unknown mechanism, activates CheY to generate clockwise rotation. Here, using quantitative selective reaction monitoring mass spectrometry and high-resolution targeted mass spectrometry, we identified K91 and K109 as the major sites whose acetylation level in vivo increases in response to acetate. Employing single and multiple lysine replacements in CheY, we found that K91 and K109 are also the sites mainly responsible for acetate-dependent clockwise generation. Furthermore, we showed that clockwise rotation is repressed when residue K91 is nonmodified, as evidenced by an increased ability of CheY to generate clockwise rotation when K91 was acetylated or replaced by specific amino acids. Using molecular dynamics simulations, we show that K91 repression is manifested in the conformational dynamics of the β4α4 loop, shifted toward an active state upon mutation. Removal of β4α4 loop repression may represent a general activation mechanism in CheY, pertaining also to the canonical phosphorylation activation pathway as suggested by crystal structures of active and inactive CheY from Thermotoga maritima. By way of elimination, we further suggest that K109 acetylation is actively involved in generating clockwise rotation.
The bacterial flagellar motor is both chemo- and mechanosensitive. It is sensitive to the intracellular concentration of the chemotaxis response regulator CheY-P and to external load conditions. The motor's dose-response curve, which represents the probability of the motor rotating clockwise (CW bias) as a function of CheY-P concentration, characterizes its chemical sensitivity.
While the basal region of the left ventricle (LV) rotates in a clockwise (cw) direction, the apical regions of the LV rotate in a counterclockwise (ccw) direction in healthy circumstances. Although LV rotational mechanics help optimize LV ejection, in some cases, LV twist is missing. This clinical situation, when the LV base and the apex rotate in the same cw or ccw direction, is called LV 'rigid body rotation' (LV-RBR).
Desmin and Vimentin are major intermediate filaments at the Z-disc and play significant roles in sarcomere architecture and signaling transduction. Abnormal expression of sarcomeric Desmin and Vimentin (SDV) results in severe dysfunctions of striated muscles. In this study, it was found that paired Numb family proteins (NFPs), including Numb and its homolog Numblike, determined the range for the recruitment of SDV to the primitive Z-disc.
Wandering spleen is a relatively rare condition and may be complicated by intestinal obstruction or abnormal intestinal rotation. Herein, we report a case where these three conditions appeared concomitantly. An 18-year-old woman with an intellectual disability was admitted to the hospital because of vomiting and fever.
In two experiments, we explored whether cross-modal cues can be used to improve foraging for multiple targets in a novel human foraging paradigm. Foraging arrays consisted of a 6 × 6 grid containing outline circles with a small dot on the circumference. Each dot rotated from a random starting location in steps of 30°, either clockwise or counterclockwise, around the circumference.
