Background: Growing evidence indicates that noncombustible products could be a tobacco harm reduction tool for smokers who do not quit. The Tobacco Heating System (THS) emits substantially lower levels of harmful cigarette smoke constituents, and previous randomized clinical studies showed improved levels of biomarkers of potential harm (BoPH) linked to smoking-related disease.

Methods: In this cross-sectional study of healthy participants (n = 982) who (i) smoked cigarettes, (ii) had voluntarily switched from smoking to THS use, or (iii) formerly smoked, blood and urine samples were assayed for nine BoPH.

In this article, we present eight studies published in the past two years that are likely to influence general practice in 2025. The key messages highlight the importance of physical activity in reducing cardiovascular risk, the effectiveness of aripiprazole for treatment-resistant depression, and the positive impact of guidelines on antibiotic use for acute cystitis. Furthermore, teleconsultation is more effective when supported by a pre-existing therapeutic relationship, extended leave reduces physician burnout, electronic cigarettes aid smoking cessation, and AI improves the management of electronic communications.

The hypothalamic neuropeptide system of orexin (hypocretin) neurons provides projections throughout the neuraxis and has been linked to sleep regulation, feeding and motivation for salient rewards including drugs of abuse. However, relatively little has been done to examine genes associated with orexin signaling and specific behavioral phenotypes in humans. Here, we tested for association of twenty-seven genes involved in orexin signaling with behavioral phenotypes in humans.
