Telavancin biological activity, determined by serum titers against a reference strain of Staphylococcus aureus, was maintained in the serum of subjects with severe renal impairment or end-stage renal disease suggesting that there is no apparent effect of renal function on in vitro activity of telavancin.

Embryonic exposure to glycopeptides induces acute toxicity and developmental neurotoxicity in zebrafish (Danio rerio).

Glycopeptide antibiotics are crucial in combating severe bacterial infections. However, concerns persist regarding their environmental impact and potential toxicity to human health. This study explores the dose-dependent toxicity of glycopeptide antibiotics on zebrafish embryos, revealing significant differences in developmental and neurological effects.

Gram-positive bacterial infections present a major clinical challenge, with methicillin- and vancomycin-resistant strains continuing to be a cause for concern. In recent years, semisynthetic vancomycin derivatives have been developed to overcome this problem as exemplified by the clinically used telavancin, which exhibits increased antibacterial potency but has also raised toxicity concerns. Thus, glycopeptide antibiotics with enhanced antibacterial activities and improved safety profiles are still necessary.

Rapid activity of telavancin against and protection against inhalation anthrax infection in the rabbit model.

Antimicrob Agents Chemother

July 2024

Product Development Division, Cumberland Pharmaceuticals, Nashville, Tennessee, USA.

Inhalation anthrax is the most severe form of infection, often progressing to fatal conditions if left untreated. While recommended antibiotics can effectively treat anthrax when promptly administered, strains engineered for antibiotic resistance could render these drugs ineffective. Telavancin, a semisynthetic lipoglycopeptide antibiotic, was evaluated in this study as a novel therapeutic against anthrax disease.

Vancomycin and daptomycin are first-line drugs for the treatment of complicated methicillin-resistant (MRSA) infections, including bacteremia. However, their effectiveness is limited not only by their resistance to each antibiotic but also by their associated resistance to both drugs. It is unknown whether novel lipoglycopeptides can overcome this associated resistance.

Purpose: The purpose of this study was to assess the potential of new lipoglycopeptides as novel topical therapies for improved treatment of recalcitrant ocular infections. We evaluated the in vitro antimicrobial activity of oritavancin, dalbavancin, and telavancin compared with vancomycin (VAN) against a large collection of ocular staphylococcal isolates and their cytotoxicity on human corneal epithelial cells (HCECs).

Methods: Antimicrobial susceptibility testing was performed by broth microdilution against 223 Staphylococcus spp.
