Enhancer of zeste homolog 2 (EZH2), an epigenetic regulator and H3k27-specific histone methyltransferase, is important for transcriptional regulation. EZH2 has been found to be overexpressed in B-cell lymphomas, as well as some T-cell lymphomas. Here we investigated the expression of EZH2 by immunohistochemical staining in a wide range of T-cell neoplasms. We found that EZH2 is highly expressed in all categories of T-cell neoplasia studied, and its expression strongly correlates with a high proliferation rate. Although up-regulation of EZH2 has been reported to be modulated by the pSTAT3-MYC pathway, our data indicate that EZH2 expression is correlated with MYC and/or pSTAT3 expression in only a subset of T-cell lymphomas, and that other mechanisms may control the overexpression of EZH2 in many T-cell lymphomas. The high level of EZH2 expression in T cell lymphomas suggest that these neoplasms may benefit from targeted treatment with a small molecule inhibitor of EZH2 currently in use in clinical trials.

Anaplastic lymphoma kinase positive (ALK+) anaplastic large cell lymphoma (ALCL) typically affects young individuals and, despite high responsiveness to cytotoxic drugs, relapses occur in over 50% of patients. Crizotinib has improved outcomes, but its management in patients desiring parenthood remains an issue. This study presents the first description of four successful pregnancies during crizotinib treatment for ALK+ALCL: a female patient achieving two pregnancies through assisted reproductive technologies (ART), temporarily discontinuing crizotinib and maintaining a complete remission (CR), and a male patient conceiving naturally while on continuous therapy.

Six pyridine analogs of (E)-3-(3-(1,2,3,4-tetrahydro-1,1,4,4,6-pentamethylnaphthalen-7-yl)-4-hydroxyphenyl)acrylic acid-or CD3254 (11)-in addition to two novel analogs of 1-(3,5,5,8,8-pentamethyl-5,6,7,8-tetrahydronaphthalen-2-yl)-1H-benzo[d][1,2,3]triazole-5-carboxylic acid (CBt-PMN or 23) were prepared and evaluated for selective retinoid-X-receptor (RXR) agonism alongside bexarotene (1), an FDA-approved drug for cutaneous T-cell lymphoma (CTCL). Treatment with 1 often elicits side-effects by disrupting or provoking other RXR-dependent nuclear receptors and cellular pathways. All analogs were assessed through modeling for their ability to bind RXR and then evaluated in human colon and kidney cells employing an RXR-RXR mammalian-2-hybrid (M2H) system and in an RXRE-controlled transcriptional assay.

Its typical characteristics include an aggressive course, progressive lymphadenopathy, hepatosplenomegaly, systemic symptoms, anemia, hypergammaglobulinemia, and generally poor prognosis.
Methods: We describe a rare case in which the left inguinal lymph node was completely excised and biopsied one year ago.
