Highly porous 45S5 Bioglass(®)-based scaffolds with interconnected pore structure are promising candidates for bone tissue engineering due to their bioactivity, biocompatibility, osteogenic and angiogenic effects. In the present study, to ensure the mechanical competence of the 45S5 Bioglass(®)-based scaffolds, their stiffness was adjusted by applying polymer coatings and further crosslinking treatment. A non-destructive ultrasonic technique was used to determine the stiffness of the scaffolds. The stiffness of uncoated scaffolds was shown to increase by applying polymer coatings, and a further increase was achieved by crosslinking the used polymer coatings. All uncoated and polymer-coated scaffolds were confirmed to exhibit stiffness values in the range of reported values in the literature for cancellous bone. A statistical evaluation of combined multiscale ultrasound-nanoindentation measurements indicated that the stiffness of the coated scaffold is directly dependent on the stiffness of the polymer coating.

This study evaluated the osteogenic potential of the bioactive glasses SinGlass (45S5) and SinGlass High (F18) in regenerating critical bone defects in rat calvaria. Both biomaterials promoted new bone formation around the particles, with the SinGlass High (F18) group exhibiting a higher rate of bone maturation. Histomorphological and birefringence analyses revealed better organization of the newly formed bone in the biomaterial-treated groups, and immunohistochemistry indicated the expression of osteogenic markers such as osteocalcin, immunostaining for bone morphogenetic protein 2 (BMP 2), and immunostaining for bone morphogenetic protein 4 (BMP 4).

Although models have been proposed to explain the mechanisms of bioglass (BG) dissolution and subsequent calcium phosphate (CaP) mineralization, open questions remain. The processes in which phase transition occurs in aqueous solutions and their dynamics remain underexplored partly because traditional instruments/techniques do not allow for direct observations at the adequate time and length scales at which such phase transformations occur. For instance, given the crucial role of the silica gel in CaP formation during BG dissolution, uncertainty exists about how such a silica gel forms on the BG surface.

This study focuses on the development of polymer-bioglass composite bone scaffolds for the treatment of bone defects. PCL particles and 45s5 bioglass powder were employed as raw materials to fabricate PCL/45s5 composite wires with mass fractions of 5 wt%, 10 wt%, and 20 wt% via the twin-screw extrusion method. A cylindrical porous model was established using 3D modeling software, and a porous composite scaffold was constructed through the melt deposition manufacturing process.
