Background: Smoking remains one of the major public health threats, necessitating substantial scientific and societal interest in further developing and implementing systematic, smoking cessation trials. This review examines ongoing randomized controlled trials (RCTs) on smoking cessation and harm reduction, focusing on adherence to German S3 guidelines for tobacco dependence and identifying areas needing further research and neglected aspects in the implementation of treatment guidelines for tobacco dependence.
Methods: A systematic search was conducted on the International Clinical Trials Registry Platform, comprising multiple trial registries worldwide, to identify ongoing RCTs focusing on smoking cessation and harm reduction.

Introduction: The objective of this systematic review was to provide an overview of current developments and potentially available therapeutic options for polymyalgia rheumatic (PMR) and giant cell arteritis (GCA), in the coming years.
Methods: We conducted a systematic review of 17 national and international clinical trial databases for all disease-modifying anti-rheumatic drugs (DMARDs) for PMR and GCA that are already marketed, in clinical development or withdrawn. The search was performed on January 2024, with the keywords "polymyalgia rheumatica" and "giant cell arteritis".

Launched in 1992, the FDA accelerated approval program grants drugs indicated in rare/life threatening diseases the ability to be marketed at a faster pace than through the traditional track. Each manufacturing company presents its drug to the FDA, and within 60 days it will determine if the drug is eligible for this path. Many drugs that were initially approved through this route, subsequently did not demonstrate their clinical benefits.
