Osmolytes have been proposed as treatments for neurodegenerative proteinopathies including Alzheimer's disease. However, for osmolytes to reach the clinic their efficacy must be improved. In this work, copper(I)-catalyzed azide-alkyne cycloaddition chemistry was used to synthesize glycoclusters bearing six copies of trehalose, lactose, galactose or glucose, with the aim of improving the potency of these osmolytes via multivalency. A trehalose glycocluster was found to be superior to monomeric trehalose in its ability to retard the formation of amyloid-beta peptide 40 (Aβ40) fibrils and protect neurons from Aβ40-induced cell death.

Enzyme-instructed signal generation at liquid-liquid interfaces presents a novel strategy for controlling and detecting biochemical processes on macroscopic scales. Here, we explore the self-assembly and jamming of pillar[5]arene (P[5]A) derivatives at the oil-water interface via a copper-mediated "click" reaction, providing a versatile platform for generating observable signals. The formation of a pillar[5]arenes network at the droplet interface reduces interfacial tension, allowing droplets to adopt various nonequilibrium shapes based on the interfacial jamming process.

This study aims to explore the development of natural bio-based amphiphilic block copolymers for drug delivery applications. We investigated block copolymers derived from tamarind seed xyloglucan and solanesol, focusing on their synthesis, structural analysis, aqueous self-assembly, and drug encapsulation. Specifically, xyloglucan hydrolysate segments with number-average degrees of polymerization (DPs) of between 8 and 44 (XOS, XMS, XMS, XMS, and XMS) were used as the hydrophilic blocks, whereas plant-sourced solanesol was selected as the hydrophobic segment.

In this study, we applied a systematic approach to establish an iterative workflow and to drive the chemical design of thermosensitive, in situ forming injectables as a function of the intended target product profile. Self-assembly, mechanical properties, physical state, and thermal transition behavior were assessed via nuclear magnetic resonance, oscillatory rheology, turbidimetry and visual inspection techniques. Thus, poly(N-isopropylacrylamide) (PNIPAM) and poly(2-alkyl-2-oxazoline)s (PAOx)s with LCSTs below body temperature were studied before and after grafting them onto azido-substituted hyaluronic acid (HA) via strain-promoted azide-alkyne cycloaddition (SPAAC).

Divergent synthesis of triazoles was achieved using newly designed platform molecules possessing azide, alkyne, and fluorosulfonyl moieties. Consecutive conjugations by the sulfur(VI) fluoride exchange and following consecutive triazole formations allowed us to prepare a wide variety of bis(triazole)s by virtue of selective transformations. One-pot triple-click assembly of easily accessible modules led to the facile synthesis of middle-molecular-weight triazoles with various functional moieties.
