We report on an all-fiber system delivering more than 100 μJ pulses with a top-hat beam output in the few nanoseconds regime at 10 kHz. The linearly polarized flattened beam is obtained thanks to a 3-mm-long single-mode microstructured fiber spliced to the amplifier's output.

Infrared matrix-assisted laser desorption electrospray ionization (IR-MALDESI) uses an infrared laser to desorb neutral biomolecules with postionization via ESI at atmospheric pressure. The Gaussian profile of the laser with conventional optics results in the heating of adjacent nonablated tissue due to the energy profile being circular. A diffractive optical element (DOE) was incorporated into the optical train to correct for this disadvantage.

MINFLUX has achieved extraordinary resolution in superresolution imaging and single fluorophore tracking. It is based on localizing single fluorophores by rapid probing with a patterned beam that features a local intensity minimum. Initially, we theoretically derive the expression of the depolarization loss in a bulk laser end-pumped with a near-field flat-top-hat or ring-shaped beam, where a significant reduction of depolarization loss in the latter case is presented. Experimental verification is thereafter carried out with a Tm:YO ceramic laser employing these two different pump configurations.
