Objective: To explore decreased proliferation ability and differentiation potential of mesenchymal stem cells (MSCs) of osteoporosis rat.
Methods: MSCs were obtained from osteoporosis rat, and proliferation potency and impaired osteogenic differentiation potential were determined.
Results: The result showed a significant downregulation of MSCs pluripotency related gene (Oct 4) and osteogenic genes (BSP, OCN) expression in OVX MSCs compared with Sham MSCs (P<0.05).
Conclusions: These data suggest that MSCs are aging in osteoporosis body, and autologous OVX MSCs transplantation is not appropriate to treat osteoporosis if necessary. There will be a possibility in establishing a new clinical application of MSCs autologous transplantation to treat osteoporosis, if OVX MSCs have stronger proliferation and differentiation.

Research Question: Can attenuated total reflection-Fourier transform infrared spectroscopy combined with machine learning techniques be used to develop a real-time diagnostic modality for chronic endometritis by analysing endometrial biopsies obtained during hysteroscopy?
Design: Women undergoing hysteroscopy for infertility assessment were enrolled in this prospective study from January 2020 to March 2021. Endometrial biopsies were evaluated using a spectrophotometer, and subsequently via histopathology, including immunohistochemical staining for the multiple myeloma oncogene-1 (MUM-1). Spectroscopy analyses of the positive and the negative chronic endometritis groups were compared across various cut-offs of MUM-1 positive cells per 10 high-power fields (HPF).

To enhance the biomarker diagnostics sensitivity and selectivity of human papillomavirus type 16 oncoprotein E7 (HPV16 E7) in serum, a label/enzyme-free electrochemical detection platform was developed. This platform featured a type of "Super-turn-off" nanobiosensor monitored through differential pulse voltammetry (DPV). It integrated the magnetic self-assembly property of the α-FeO/FeO@Au/Sub/BSA signal transport nano-medium with the high specificity of CRISPR/Cas14a and the amplification capability of the bipedal walker (DNA walker composed of two ssDNA strands), resulting in the enhanced specificity and anti-interference performance while remaining stable at 4 °C for over 30 days.

X-linked ichthyosis (XLI) is a genetic disorder characterized by a steroid sulfatase (STS) deficiency inducing excessive cholesterol sulfate accumulation and keratinization. Our study utilizes STS knockout mice to reproduce the hyperkeratinization typical of XLI, providing a valuable model for investigating the underlying mechanisms. From the experiment of STS-deficient keratinocytes using the CRISPR/Cas9 system, we observed upregulation of E-cadherin, which is associated with keratinocyte differentiation and stratification.
