The formation of multilamellar vesicles (MLVs) from the lamellar phase of nonionic surfactant system C12E5/D2O under shear flow is studied by time-resolved small angle neutron and light scattering during shear flow. A novel small angle neutron scattering sample environment enables the tracking of the lamellae alignment in the velocity-velocity gradient (1-2) plane during MLV formation, which was tracked independently using flow small angle light scattering commensurate with rheology. During the lamellar-to-multilamellar vesicle transition, the primary Bragg peak from the lamellar ordering was observed to tilt, and this gradually increased with time, leading to an anisotropic pattern with a primary axis oriented at ∼25° relative to the flow direction. This distorted pattern persists under flow after MLV formation. A critical strain and critical capillary number based on the MLV viscosity are demonstrated for MLV formation, which is shown to be robust for other systems as well. These novel measurements provide fundamentally new information about the flow orientation of lamellae in the plane of flow that cannot be anticipated from the large body of previous literature showing nearly isotropic orientation in the 2,3 and 1,3 planes of flow. These observations are consistent with models for buckling-induced MLV formation but suggest that the instability is three-dimensional, thereby identifying the mechanism of MLV formation in simple shear flow.

Objectives: To investigate the pathogenicity of feline herpesvirus-1 (FHV-1) to the cornea, FHV-1 strains isolated from feline eyes with dendritic ulcers were subjected to genomic analysis to determine whether FHV-1 vaccine strains are involved in the formation of dendritic ulcers.
Methods: All open reading frame (ORF) sequences of the three F2 strains (Virbac, Intervet and Merial) and the FHV-1 clinical isolates from cats registered in GenBank were compared to detect nucleotide variants unique to the F2 strains, with those nucleotides then being used for simple genotyping of the F2 strains. In all isolates from feline eyes with dendritic ulcers, the regions including nucleotide variants of the F2 strain were amplified with PCR and sequenced.

Formation of unilamellae of fully hydrated dipalmitoylphosphatidylcholine (DPPC) was induced by a horn sonicator from multilamellar vesicles and followed by time-resolved synchrotron small angle X-ray scattering and direct visual morphological investigations by the means of transmission electron-microscopy combined with freeze-fracture. Without incubation the ultrasonication causes continuous increasing in temperature and transformation from the gel to rippled gel structures, then reaching the main transition, the formfactor of unilamellar structure appeared. The ultrasonication resulted in different layer formations at the characteristic temperatures of the gel (20 °C), rippled gel (38 °C), and liquid crystalline (45 °C) phases of the system.

Nanosized liposomal vesicles (NLV) were successfully prepared using natural sunflower lecithin without the use of high-pressure homogenization or filtration. Upon glycerol addition to dispersions of lecithin multilamellar vesicles (MLVs), these broke down spontaneously to liposomes with diameters in the range of 100-200 nm. Static light scattering demonstrated that glycerol addition above 30% (w/w) induced the complete transformation of MLVs into NLVs.
