Nanomaterials have made a significant impact on cancer therapeutics and an emergence of polymeric nanoparticle provides a unique platform for delivery of drug molecules of diverse nature. Nanoparticles can be targeted at the tumor cells due to enhanced permeability and retention effect. Moreover, nanoparticles can be grafted by various ligands on their surface to target the specific receptors overexpressed by cancer cells or angiogenic endothelial cells. These approaches ultimately result in longer circulation half-lives, improved drug pharmacokinetics, reduced side effects of therapeutically active substances and overcoming cancer chemo-resistance thereby enhancing the therapeutic efficacy of the treatment. This review article summarizes the recent efforts in cancer nanochemotherapeutics using polymeric nanoparticles with a special reference to their pharmacokinetic and biodistribution profiles, their role in reversing multidrug resistance in cancer and strategies of tumor targeting with them, along with the challenges in the field.

An electro- and optically favorable quaternary nanocomposite film was produced by solution-casting nickel oxide nanoparticles (NiO NPs) into polyvinyl alcohol (PVA), polyvinyl pyrrolidone (PVP), and poly(3,4-ethylenedioxythiophene) polystyrene sulfonate (PEDOT/PSS). Based on transmission electron microscopy (TEM) and X-ray diffraction (XRD) observations, the synthesized NiO NPs have a cubic phase and a diameter between 10 and 45 nm. The complexity and interactions observed through XRD patterns, UV-visible spectra, and FTIR measurements suggest that the NPs are not just dispersed within the polymer matrix, but are interacting with it, leading to enhanced dielectric properties and AC electrical conductivity.

Functionalization of polymer nanoparticles (NPs) with targeting peptides is of interest for drug delivery applications to enhance tumor accumulation and penetration. Herein, we evaluated the feasibility of two different methods for the attachment of a tumor-penetrating peptide LinTT1 (AKRGARSTA) to poly(ethylene glycol)-block-poly(ε-caprolactone) (PCL-PEG) NPs: (1) "post-conjugation" onto pre-formed nanoparticles, and (2) "pre-conjugation", the synthesis and purification of peptide-polymer conjugates and subsequent nanoprecipitation of the conjugates diluted with non-functionalized polymers. Conjugation of the labelled peptide via maleimide-thiol chemistry was verified by gel permeation chromatography (GPC) and fluorescence measurements.
