The formation of atomically precise covalent networks directly on a surface is a promising approach to produce single layers of two-dimensional organic materials (2D polymers). In the emerging field of on-surface synthesis, there is an urgent need for finding a rationale to new reaction pathways taking place directly on the surface. In this feature article we review and put into perspective recent results on the surface polymerisation of boronic acid based systems under ultrahigh vacuum conditions studied by scanning tunnelling microscopy. By presenting several approaches to control the growth of covalent networks, we aim at providing a comprehensive overview of the complex mechanisms involved.

Three cases of aminobenzoic acid hybrid polyoxotungstates, Na(HO)[(HPWO) (OCCHNH)]·7HO (), K(HO)[(AsWO)(OCCHNH)]·4HO (), and [(HN(CH)]Na(HO)[(SbWO) (OCCHNH)]·7HO (), were successfully synthesized. This is the first report of the successful assembly of the hexanuclear {XW} (X = HP, As, or Sb) clusters and organic carboxylic acid (para aminobenzoic acid) ligands. All three hybrids feature a common {XW} unit composed of a six-membered {WO} octahedral ring capped by one {XO} trigonal pyramid.

Proximity labeling (PL) has emerged as a powerful technique for the in situ elucidation of biomolecular interaction networks. However, PL methods generally rely on single-biological-hierarchy control of spatial localization at the labeling site, which limits their application in multi-tiered biological systems. Here, we introduced another enzymatic reaction upstream of an enzyme-based PL reaction and targeted the two enzymes to markers indicating different biological hierarchies, establishing a two-level spatially localized proximity labeling (P2L) platform for in situ molecular measurement and manipulation.

Template-directed synthesis has become a powerful methodology to access complex molecules. Noncovalent templating has been widely used in the last few decades, but less attention has been paid to covalent template-directed synthesis, despite the fact that this methodology was used for the first reported synthesis of a catenane. This review highlights the evolution of covalent templating over the last 60 years, thereby providing a toolbox for the design of efficient covalent templating processes.
