Light-mediated control of gene expression in filamentous fungus Trichoderma reesei.

We developed a light-mediated system based on synthetic light-switchable transactivators. The transactivators bind promoter upon blue-light exposure and rapidly initiate transcription of target transgenes in filamentous fungus Trichoderma reesei. Light is inexpensive to apply, easily delivered, and instantly removed, and thus has significant advantages over chemical inducers.

In Singapore's hot and humid climate, watercolor papers are particularly prone to a paper oxidation issue known as foxing, which refers to the discoloration forming yellowish-brown stains on paper, changing the visual outcome of the watercolor artworks. This research investigates two most popular types of watercolor paper, made from 100% cotton and cotton-wood-pulp mixture. Foxing was generally categorized into two types: biotic and abiotic foxing caused by fungi activities and the presence of metallic contaminants catalytic fungi growth.

Three previously undescribed indole diketopiperazine alkaloids and seventeen known compounds were characterized from Aspergillus sp. JXC-5 by solid fermentation. Their structures were elucidated by spectroscopic methods and HRESIMS, and the absolute configurations were further confirmed by electronic circular dichroism (ECD), induced CD spectra and ML_J_DP4 methods.

The discovery of novel natural products through the exploration of distinct microorganisms is crucial for advancing drug discovery research. In this study, we focus on a unique environmental resource, microbial masses known as "Tengu-no-Mugimeshi."
