Chromatographic purification of the methanolic extract from the soft coral Sarcophyton pauciplicatum led to the isolation of three polyhydroxylated steroids 1-3, including a new compound, sarcopanol A (1). Their structures were elucidated by spectroscopic analysis and by comparison of the spectroscopic data with those of similar compounds previously reported in literature. The anti-inflammatory effects of isolated compounds were evaluated using nuclear factor kappa B (NF-κB) luciferase and reverse transcription polymerase chain reaction (RT-PCR). The effect of isolated compounds on cell growth was evaluated by MTS assays. Compounds 1 and 2 significantly inhibited TNFα/INFγ-induced NF-κB transcriptional activity in human keratinocyte (HaCaT) cells in a dose-dependent manner, with EC50 values of 8.27±3.28 and 26.07±5.59 μM, respectively. Furthermore, the transcriptional inhibition of these compounds was confirmed by a decrease in cyclooxygenase-2 (COX-2), inducible nitric oxide synthase (iNOS), and intercellular adhesion molecule-1 (ICAM-1) gene expression levels in HaCaT cells.

Two undescribed rearranged diterpenoids, lobopaucinoids A and B (1 and 2), along with thirteen undescribed lobane-type diterpenoids lobopaucinoids C-O (3-15) including a C (11) and two C (12 and 13) undescribed norditerpenoids, were isolated from the soft coral Lobophytum pauciflorum Ehrenberg (Sarcophytidae family) collected from Xisha Islands of the South China Sea. Additionally, two undescribed prenyleudesmane-type diterpenoids, lobopaucinoids P and Q (16 and 17), as well as two known lobane diterpenoids (18 and 19), were also obtained. Aims: In the present study, we tested if terrestrially-derived humic substances (HS) could mitigate the adverse effects of elevated temperature and UVB radiation on the bacterial communities of two hard corals (Montipora digitata and Montipora capricornis), one soft coral (Sarcophyton glaucum), sediment and water. We also examined the impact of temperature, UVB radiation and HS supplementation on coral photosynthetic activity, a proxy for coral bleaching.

New technology has opened opportunities for research and exploration of deep-water ecosystems, highlighting deep-sea coral reefs as a rich source of novel bioactive natural products. During our ongoing investigation of the chemodiversity of the Irish deep sea and the soft coral we report 12 unreported cadinene-like functionalized sesquiterpenes, anthoteibinenes F-Q. The metabolites were isolated using both bioassay- and H NMR-guided approaches.
