The mulberry white caterpillar, Rondotia menciana, belongs to the lepidopteran family Bombycidae, in which the domestic silkworm, Bombyx mori is included. In this study, we describe the complete mitochondrial genome of R. menciana in terms of general genomic features and characteristic features found in the A+T-rich region. The 15,364 bp long genome consisted of a typical set of genes (13 protein-coding genes [PCGs], 2 rRNA genes, and 22 tRNA genes) and 1 major non-coding A+T-rich region, with the typical arrangement found in Lepidoptera. Twelve of the 13 PCGs started with typical ATN codons, except for the COI, which began with CGA and twelve of 13 PCGs had complete stop codons, except for the COII, which ended with a single T. The 360 bp long A+T-rich region harbored the conserved sequence blocks typically found in lepidopteran insects. Additionally, the A+T-rich region of R. menciana contained one tRNA(Met)-like structure, which had a proper anticodon and secondary structure.

Using next-generation sequencing data, the complete mitogenomes of six species from the genus were assembled. This study explores the mitochondrial genomes of species, among them the five species from the complex, comparing them with each other and with other species from Dolichoderinae subfamily to understand their evolutionary relationships and evolution. mitochondrial genomes contain the typical set of 13 protein-coding genes, two ribosomal RNA genes, 22 transfer RNAs, and the A + T-rich control region.

The chloride transporter-channel SLC26A9 is mediated by a reciprocal regulatory mechanism through the interaction between its cytoplasmic STAS domain and the R domain of CFTR. In vertebrate Slc26a9s, the STAS domain structures are interrupted by a disordered loop which is conserved in mammals but is variable in non-mammals. Despite the numerous studies involving the STAS domains in SLC26 proteins, the role of the disordered loop region has not been identified.

Previous studies on the gene order and composition of leafhopper mitochondrial genomes have revealed a high level of conservation in overall genome structure. Some members of Deltocephalinae, the largest subfamily, exhibit tRNA gene rearrangements; however, few mitogenomes have been sequenced in this group and the degree of structural variation within tribes remains unclear. In this study, we sequenced the complete mitogenomes of 14 species belonging to four genera of tribe Deltocephalini from China and compared them with the two previously reported mitogenomes for this tribe.
