Objective: To investigate whether the magnitude of presynaptic dopamine depletion is a risk factor for the development of levodopa-induced dyskinesia (LID) in Parkinson disease (PD) by quantitatively analyzing (18)F-FP-CIT PET data.
Methods: This retrospective cohort study enrolled a total of 127 drug-naive de novo patients with PD who completed (18)F-FP-CIT PET scanning at their initial evaluation. The patients visited our outpatient clinic every 3-6 months and had been followed for a minimum of 2 years since beginning dopaminergic medication. The predictive power of the quantitatively analyzed (18)F-FP-CIT uptake of striatal subregions and other clinical factors for the development of LID was evaluated using Cox proportional hazard models.
Results: During a mean follow-up period of 3.4 years, 35 patients with PD (27.6%) developed LID. Patients with LID showed less dopamine transporter (DAT) activity in the putamen than did those without LID. Multivariate Cox proportional hazard models revealed that the DAT uptakes of the anterior putamen (hazard ratio [HR] 0.530; p = 0.032), posterior putamen (HR 0.302; p = 0.024), and whole putamen (HR 0.386; p = 0.022) were significant predictors of the development of LID, whereas DAT activities in the caudate and ventral striatum were not significantly correlated with the development of LID. In addition, younger age at onset of PD and higher dose of levodopa were also significant predictors of the development of LID.
Conclusions: The present results provide convincing evidence that presynaptic dopaminergic denervation in PD plays a crucial role in the development of LID.

Purpose: The primary objective of this study was to investigate the possible role and assess the potential relationship of speculum use during phacoemulsification surgery in the occurrence of blepharoptosis.
Methods: Our study is a prospective observational design to analyze patients who underwent phacoemulsification surgery under topical anesthesia between October 2017 and May 2018 at a tertiary eye hospital in South India. All patients had their Margin Reflex Distance 1 (MRD1), levator palpebrae superioris (LPS) function, and lid crease distance (LCD) measured before and after surgery on day one, one month, three months, and six months to evaluate the extent of ptosis at each time point that was recorded clinically and photographic documentation for accurate evaluation.
CRISPR-Cas12a is widely used for genome editing and biomarker detection since it can create targeted double-stranded DNA breaks and promote non-specific DNA cleavage after identifying specific DNA. To mitigate the off-target DNA cleavage of Cas12a, we previously developed a Cas12a variant (FnoCas12a ) by introducing double proline substitutions (K969P/D970P) in a conserved helix called the bridge helix (BH). In this work, we used cryogenic electron microscopy (cryoEM) to understand the molecular mechanisms of BH- mediated activation of Cas12a.

The comprehensive benefit evaluation of LID based on multi-criteria decision-making methods faces technical issues such as the uncertainties and vagueness in hybrid information sources, which can affect the overall evaluation results and ranking of alternatives. This study introduces a multi-indicator fuzzy comprehensive benefit evaluation approach for the selection of LID measures, aiming to provide a robust and holistic framework for evaluating their benefits at the community level. The proposed methodology integrates quantitative environmental and economic indicators with qualitative social benefit indicators, combining the use of the Storm Water Management Model (SWMM) and ArcGIS for scenario-based analysis, and the use of hesitant fuzzy language sets and Technique for Order of Preference by Similarity to Ideal Solution (TOPSIS) for decision-making.

Background: The paramedian forehead flap (PMFF) has been well established for use in reconstruction limited to a single nasal outer layer defect and has recently gained recognition as an acceptable alternative to traditional methods of lower eyelid reconstruction. The use of a single, pedicled PMFF for the reconstruction of more than one defect has yet to be described.
Methods: A 59-year-old male patient was originally diagnosed with large squamous cell carcinoma resulting in radical resection and ipsilateral neck dissection.

Urban flooding poses a significant risk to cities worldwide, exacerbated by increasing urbanization and climate change. Effective flood risk management requires comprehensive assessments considering the complex interaction of social, economic, and environmental factors. This study developed an innovative Urban Flood Risk Index (FRI) to quantify and assess flood risk at the sub-catchment level, providing a tool for evidence-based planning and resilient infrastructure development.
