Recent clinical studies have demonstrated that dual orexin receptor antagonists (OX1R and OX2R antagonists or DORAs) represent a novel treatment option for insomnia patients. Previously we have disclosed several compounds in the diazepane amide DORA series with excellent potency and both preclinical and clinical sleep efficacy. Additional SAR studies in this series were enabled by the expansion of the acetonitrile-assisted, diphosgene-mediated 2,4-dichloropyrimidine synthesis to novel substrates providing an array of Western heterocycles. These heterocycles were utilized to synthesize analogs in short order with high levels of potency on orexin 1 and orexin 2 receptors as well as in vivo sleep efficacy in the rat.

Background: The orexin/hypocretin neuropeptide system, primarily found in the lateral hypothalamus and perifornical region, modulates sleep, wakefulness, appetite, and cognitive function. One region with dense orexinergic projections is the basal forebrain (BF), which is the major source of acetylcholine in the neocortex and limbic structures such as the hippocampus. The basal forebrain cholinergic system mediates cognition and dysfunction is one of the key hallmarks of Alzheimer's disease.

December 2024

Research Program on Cognition & Neuromodulation Based Interventions, Ann Arbor, MI, USA.

Background: The multisite SuperAging Research Initiative (SRI) was established in 2021 to identify resilience and resistance factors promoting cognitive healthspan through a harmonized multidisciplinary protocol with prospective data collection. The designation of SuperAger is reserved for individuals age 80+ with episodic memory performance that is at least average for those 2-3 decades younger. Research studies of this relatively uncommon phenotype allow for investigations of fundamental importance to the neurobiology of brain aging, resilience, resistance, and avoidance of cognitive decline related to "average aging" and more severe impairments associated with Alzheimer's and related dementias (ADRD).

Background: Disruption of sleep and circadian rhythms are associated with cognitive decline, preclinical Alzheimer's Disease (AD) pathology, and increased risk of dementia. Alleviating circadian rhythm and sleep disruption may improve cognition and reduce the progression of AD and related dementias (ADRD). Time-restricted eating (TRE), a circadian behavioral intervention that corrects disrupted eating rhythms by aligning food intake to the daytime, has demonstrated improvements in metabolic dysfunction and sleep quality.

Omega-3 fatty acids and sleep: recent advances in understanding effects and mechanisms.

Curr Opin Clin Nutr Metab Care

December 2024

Brain, Performance and Nutrition Research Centre, Faculty of Health and Life Sciences, Northumbria University, UK.

Purpose Of Review: Sleep disturbance and low dietary intake of omega-3 fatty acids are common in modern society. Previous research shows that omega-3 fatty acids play an integral role in the regulation of sleep processes, but efficacy data in adults have been equivocal to date. This review examines recent studies highlighting the effects of omega-3 fatty acids on sleep quality and circadian processes.
