Chromosome damage in vitro after bleomycin treatment during the late S and G2 phases of the cell cycle was studied in the peripheral lymphocytes of 19 untreated patients with primary testicular tumours and 22 age-matched healthy men with no excess of cancer incidence in the families. The occurrence of spontaneous chromosome aberrations was not shown to be different in the studied groups. However, in the lymphocytes treated with bleomycin, cancer patients exhibited higher numbers of break events per cell (1.06 versus 0.67, P less than 0.01) and increased frequency of cells with aberrations (55.0 versus 43.0, P less than 0.05) than control group. Aberrant cells of cancer patients had more aberrations than cells of the control sample (1.79 versus 1.53, P less than 0.01). The frequency of chromosome 1 aberrations, often encountered in cancer cells of testicular and other solid tumours, was significantly higher in lymphocytes of patients with testicular cancer (15.0 versus 8.4%, P less than 0.0001), the long arm of this chromosome being predominantly affected (12.0 versus 6.3%, P less than 0.0001). These results support the view that a genome disposed to testicular cancer is less effective in the ability to repair non-specific DNA damage in this region, more susceptible to damage, or both.

Testicular cryopreservation has been highlighted as a promising alternative for preserving male fertility and can be applied to restore spermatogenesis in prepubertal individuals or cancer patients, preserve biologically valuable genotypes, and in studies on reproductive physiology or toxicity of various substances. This review presents an analysis of the technical aspects and applications of testicular cryopreservation, examining the contributions of important studies in this area and discussing the different factors that can impact the efficiency of the technique. Testicular fragments can be obtained from living or dead individuals, at any age and reproductive stage, through orchiectomy or biopsy.

Testicular germ cell tumour (TGCT) is a malignancy with known inherited risk factors, affecting young men. We have previously identified several hundred differentially abundant circulating RNAs in pre-diagnostic serum from TGCT cases compared to healthy controls. In this study, we performed Weighted Gene Co-expression Network Analysis (WGCNA) on mRNA and miRNA data from these samples.

Familial History in Ovarian Teratomas Is a Frequent Event: 22 Years' Experience at a Single Center.

Pediatr Blood Cancer

January 2025

Department of Pediatric Surgery, Urology and Transplantation, Hôpital Universitaire Necker-Enfants Malades, APHP, Université de Paris Cité, Paris, France.

Background: Ovarian mature teratoma represents the most common benign neoplasm among pediatric germ cell tumors. This study reports the prevalence and characteristics of familial forms identified in a single center over 22 years in order to better understand possible familial predispositions to ovarian teratoma.

Methods: The records of all patients who were surgically treated for ovarian teratoma between 2000 and 2022 were retrospectively reviewed.

Mutations in the mitochondrial (mt) genome contribute to metabolic dysfunction and their accumulation relates to disease progression and resistance development in cancer cells. This study explores the mutational status of the mt genome of cisplatin-resistant -sensitive testicular germ cell tumor (TGCT) cells and explores its association with their respiration parameters, expression of respiratory genes, and preferences for metabolic pathways to reveal new markers of therapy resistance in TGCTs. Using Illumina sequencing with Twist Enrichment Panel, the mutations of mt genomes of sensitive 2102EP, H12.
