A wide range of immunoreactive antigens commonly investigated in diagnostic pathology were evaluated in tissues subjected to fixation by microwave irradiation in normal saline to 63 degrees C. All of the 23 antigens studied were well preserved and compared with corresponding tissues conventionally fixed by formaldehyde for periods of 16 and 18 h, respectively, immunostaining in microwave-irradiated tissues was clearly superior, being more intense and also more extensive. Proteolytic digestion was not necessary for irradiated tissues except for the staining of cytokeratins and desmin. It was also possible to stain for Leu-14 (CD22) and Leu-1 (CD5), whereas these lymphocyte antigens were lost during formaldehyde fixation. Cytomorphological details were well preserved in the rapidly-fixed irradiated tissues.

The aim of the current study is to synthesize nanosized silicon incorporated HAp (Si-HAP) using sodium metasilicate as the silicon source. Previously, we, and others, have demonstrated a rapid and significant post-mortem increase in brain prostanoid (PG) levels analyzed without microwave fixation, and this is not the result of PG trapping or destruction in microwave-irradiated brain tissue. In the present study, we demonstrate a dramatic increase in kidney eicosanoid levels when analyzed without microwave fixation which was mainly accounted for by the 142-, 81-, and 62-fold increase in medullary 6-ketoPGF1α, PGE2, and PGF2α, levels, respectively, while PGD2 and TXB2 levels were increased ~7-fold. Whole kidney and cortex PG were also significantly increased in non-microwaved tissue, but at lesser extent.

Rapid inactivation of metabolism is essential for accurately determining the concentrations of metabolic intermediates in the in vivo state. We compared a broad spectrum of energetic intermediate metabolites and neurotransmitters in brains obtained by microwave irradiation to those obtained by freeze blowing, the most rapid method of extracting and freezing rat brain. The concentrations of many intermediates, cytosolic free NAD(P)(+) /NAD(P)H ratios, as well as neurotransmitters were not affected by the microwave procedure.

A rapid, simple and solvent-free procedure was developed for the determination of synthetic polycyclic musks in oyster samples by using one-step microwave-assisted headspace solid-phase microextraction (MA-HS-SPME) and gas chromatography-mass spectrometry (GC-MS). Two commonly used synthetic polycyclic musks, galaxolide (HHCB) and tonalide (AHTN), were selected in the method development and validation. The parameters (microwave irradiation power, extraction time, amount of water added, pH value and addition of NaCl) affecting the extraction efficiency of analytes from oyster slurry were systematically investigated and optimised.
