Hepatoprotective effects of triterpenoids from Ganoderma cochlear.

Two novel trinorlanostanes, cochlates A and B (1 and 2), with a 3,4-seco-9,10-seco-9,19-cyclo skeleton, as well as six new triterpenoids, fornicatins D-F (3-5) and ganodercochlearins A-C (6-8), together with five known triterpenoids (9-13), were obtained from the fruiting bodies of Ganoderma cochlear. The structural elucidation was achieved by interpretation of spectroscopic data, and compounds 2 and 7a were further characterized by X-ray crystallographic analysis. Fornicatins A, D, and F (10, 3, and 5) and fredelin (13) lowered the ALT and AST levels in HepG2 cells treated with H2O2, suggesting that they could display in vivo hepatoprotective activities.

NMR (H and C NMR, H - H COSY, HSQC, HMBC and ROESY), and HRESIMS were employed for the structure elucidation of new compounds. Four novel epimeric meroterpenoids, ganadone A (1), 3',10'-di-epi-ganadone A (2), 10'-epi-ganadone A (3), and 3'-epi-ganadone A (4) as well as another pairs of epimers, ganadone B (5) and 10'-epi-ganadone B (6), with a same basic skeleton compound ganadone C (7), together with two lactonized meroterpenoids, ganadones D and E (8 and 9) were isolated from the fruiting bodies of Ganoderma cochlear. Compounds 1-7 were constructed with fascinating adjacent 6',7'-bifuran ring system. Fortunately, we have revised our previously reported structure cochlearol Q, which was proposed pyrano[6',7'-b]pyran ring system into 6',7'-bifuran motif.

The structural elucidation of new compounds, including their absolute configurations, depends on spectroscopic analysis and electronic circular dichroism (ECD) calculations. Biological studies showed that (+)- and (-)-, (-)-, and (+)- and (-)- are cytotoxic toward human triple negative breast cancer (TNBC) cells (MDA-MB-231) with IC values of 28.
