Ribosomes are key macromolecular protein synthesis machineries in the cell. Human ribosomes have so far not been studied to atomic resolution because of their particularly complex structure as compared with other eukaryotic or prokaryotic ribosomes, and they are difficult to prepare to high homogeneity, which is a key requisite for high-resolution structural work. We established a purification protocol for human 80S ribosomes isolated from HeLa cells that allows obtaining large quantities of homogenous samples as characterized by biophysical methods using analytical ultracentrifugation and multiangle laser light scattering. Samples prepared under different conditions were characterized by direct single particle imaging using cryo electron microscopy, which helped optimizing the preparation protocol. From a small data set, a 3D reconstruction at subnanometric resolution was obtained showing all prominent structural features of the human ribosome, and revealing a salt concentration dependence of the presence of the exit site tRNA, which we show is critical for obtaining crystals. With these well-characterized samples first human 80S ribosome crystals were obtained from several crystallization conditions in capillaries and sitting drops, which diffract to 26 Å resolution at cryo temperatures and for which the crystallographic parameters were determined, paving the way for future high-resolution work.

Co-existing neuropathological comorbidities have been repeatedly reported to be extremely common in subjects dying with dementia due to Alzheimer disease. As these are likely to be additive to cognitive impairment, and may not be affected by molecularly-specific AD therapeutics, they may cause significant inter-individual response heterogeneity amongst subjects in AD clinical trials. Furthermore, while originally noted for the oldest old, recent reports have now documented high neuropathological comorbidity prevalences in younger old AD subjects, who are more likely to be included in clinical trials.

The increasing use of immune checkpoint inhibitors in clinical practice is associated with a broad spectrum of immune-related adverse events, such as cardiac, rheumatological and neurological toxicities. Myocarditis is a life-threatening complication, and the concurrent development of myocarditis, myositis and/or myasthenia leads to difficulties in diagnosis, management and treatment. We describe a case presenting with this triple M overlap syndrome following pembrolizumab treatment.

Aim: The aim of this study was to identify opportunities for more effective measures. We analyzed sex-, age- and season-specific trends in unintentional accidental deaths from stumbling falls, bathtub drowning, food choking and traffic injuries among older adults in Japan.
Methods: Death rates from major unintentional accidents were analyzed by sex, month and 5-year age groups (aged ≥50 years) based on a custom-made aggregation of data from the Vital Statistics survey (2008-2022) in compliance with the Statistics Act.

Objectives: To evaluate the multifunctionality of silver-copper co-loaded mesoporous bioactive glass (MBG), with the goal of developing an advanced pulp-capping material.
Methods: The synthesis of materials was conducted using the sol-gel method, following the approach described in previous studies but with some modifications. The composition included 80 mol% SiO₂, 15 mol% CaO, and 5 mol% P₂O₅, with additional components of 5 mol% silver, 5 mol% copper, or 1 mol% silver combined with 4 mol% copper, designated as Ag5/80S, Cu5/80S, or Ag1Cu4/80S, respectively.
