SELMA (SELection with Modified Aptamers) is a directed evolution method which can be used to develop DNA-supported clusters of carbohydrates in which the geometry of clustering is optimized for strong recognition by a lectin of interest. Herein, we report a modification of SELMA which results in glycoclusters which achieve dramatically stronger target recognition (100-fold) with dramatically fewer glycans (2-3-fold). Our first applications of SELMA yielded clusters of 5-10 oligomannose glycans which were recognized by broadly neutralizing HIV antibody 2G12 with moderate affinities (150-500 nM Kd's). In the present manuscript, we report glycoclusters containing just 3-4 glycans, which are recognized by 2G12 with Kd's as low as 1.7 nM. These glycoclusters are recognized by 2G12 as tightly as is the HIV envelope protein gp120, and they are the first constructs to achieve this tight recognition with the minimal number of Man9 units (3-4) necessary to occupy the binding sites on 2G12. They are thus of great interest as immunogens which might elicit broadly neutralizing antibodies against HIV.

Mannan and β-(1→3)-glucan are two polysaccharide markers that are characteristic for a number of fungal pathogens, including , which is the most common cause of invasive mycoses in humans. In this study, we examined epitope specificity of two monoclonal antibodies, CM532 and FG70, which recognize certain oligosaccharide fragments of these fungal polysaccharides. Using a panel of biotinylated oligosaccharides as coating antigens, we found that the CM532 antibody obtained by immunization with the pentamannoside β-Man-(1→2)-β-Man-(1→2)-α-Man-(1→2)-α-Man-(1→2)-α-Man KLH conjugate, selectively recognizes the trisaccharide β-Man-(1→2)-α-Man-(1→2)-α-Man epitope.

Metal-organic frameworks (MOFs) have become a highly usable system in various sectors because of their highly ordered structure and high porosity providing them with high storage capacity. However, their use is sometimes forbidden in the food industry due to the presence of some organic compounds which have undesirable effects. Cyclodextrins, which are considered GRAS (Generally Recognized as Safe) by the FDA, comes as a very good alternative to previously used compounds for the development of the MOFs to be used in the food packaging industry, especially in the packaging sector.
