Host⋅guest complexes between cucurbit[7] (CB[7]) or CB[8] and diamantane diammonium ion guests 3 or 6 were studied by (1) H NMR spectroscopy and X-ray crystallography. (1) H NMR competition experiments revealed that CB[7]⋅6 is among the tightest monovalent non-covalent complexes ever reported in water with Ka =7.2×10(17) M(-1) in pure D2 O and 1.9×10(15) M(-1) in D2 O buffered with NaO2 CCD3 (50 mM). The crystal structure of CB[7]⋅6 allowed us to identify some of the structural features responsible for the ultratight binding, including the distance between the NMe3 (+) groups of 6 (7.78 Å), which allows it to establish 14 optimal ion-dipole interactions with CB[7], the complementarity of the convex van der Waals surface contours of 6 with the corresponding concave surfaces of CB[7], desolvation of the CO portals within the CB[7]⋅6 complex, and the co-linearity of the C7 axis of CB[7] with the N(+) ⋅⋅⋅N(+) line in 6. This work further blurs the lines of distinction between natural and synthetic receptors.

Macrocycles that encapsulate two guests can self-sort those into homo- and heterodimers. We report here a family of self-sorting homobimetallic Pt(II) terpyridyl acetylide dimers secured together with a pair of Cucurbit[8]uril macrocycles (CB[8]). The rigid bridging unit between both Pt centers introduces varying "hinge" angles, resulting in disparities in Pt-Pt distances in the heterodimers, and leads to recognition motif mismatch.

We present the complete sequence of the genomic RNA of an isolate of squash vein yellowing virus () from California (SqVYV-CA) and show it is a recombinant virus with a highly divergent 5' UTR and proximal P1a gene. The evolution of SqVYV-CA involved an intrageneric event between unknown potyviruses, related to isolates of papaya ringspot virus () from the Old World, and an intergeneric event between this recombinant potyvirus (minor parent) and an isolate of SqVYV from Israel (SqVYV-IL) (major parent). These events occurred in mixed infections and in the potyvirus P1 and ipomovirus P1a recombination hotspots and resulted in SqVYV-CA having a potyvirus 5' UTR and chimeric P1-P1a gene/protein and the remainder of the genome from SqVYV-IL.

Methods: A hybrid strategy with Illumina and Oxford Nanopore Technology sequencing data was used to assemble bottle gourd mitochondrial and chloroplast genomes.

Gene expression technology has become an indispensable tool for elucidating biological processes and developing biotechnology. Cell-free gene expression (CFE) systems offer a fundamental platform for gene expression-based technology, in which the reversible and programmable control of transcription can expand its use in synthetic biology and medicine. This study shows that CFE can be controlled via the host-guest interaction of cucurbit[7]uril (CB[7]) with -guest-modified adenosines.

In an effort to target polypeptides at nonterminal sites, we screened the binding of the synthetic receptor cucurbit[8]uril (Q8) to a small library of tetrapeptides, each containing a nonterminal dipeptide binding site. The resulting leads were characterized in detail using a combination of isothermal titration calorimetry, H NMR spectroscopy, electrospray ionization time-of-flight mass spectrometry (ESI-TOF-MS), and X-ray crystallography. The equilibrium dissociation constant values determined for the binding of Q8 to nonterminal dipeptide sites Lys-Phe (KF) and Phe-Lys (FK) were 60 and 86 nm, respectively.
