A fluorescein-based sensor was developed for the AChE activity assay and the inhibitor screening. The sensor provided the dual assay methods for the screening of AChE activity in the presence or absence of inhibitor. The colorimetric and fluorometric assays were based on the following processes: (1) owing to the hydrolysis of acetylthiocholine in the presence of AChE, the fluorescein-based probe can rapidly induce 1,4-addition of the hydrolysis product thiocholine to α,β-unsaturated ketone in the compound 1, resulting in strong fluorescence and absorption changes; (2) in the presence of the corresponding inhibitor, the fluorescence enhancement or the absorption change would be inhibited in that the formation of thiocholine was hindered.

In this work, the interaction behaviour of gold nanoparticles (AuNPs) with o-phenylenediamine (OPD) was studied to ascertain the nanozyme-substrate interaction. The UV-Vis absorption, high-resolution transmission electron microscopy and zeta potential analysis revealed that the electron-rich nitrogen atoms in OPD showed a stronger affinity toward electron-deficient surface, indicating a stronger interaction between nanozyme and substrate molecules. Subsequently, under optimum conditions, AuNPs are used as nanozyme to catalyze the oxidation of OPD in the presence of HO.

The DPE-based chromophores exhibit a large Stokes shift and maintain good fluorescence quantum yields. A novel probe NIR8 decorated with colorimetric, fluorometric, turn-on, and NIR properties was designed and synthesized using the chromenylium-cyanine platform for sensitive and selective quantification of Hg in real samples. The interaction between the probe and Hg resulted in the opening of the closed spirolactam ring of the chromenylium-cyanine platform, leading to a notable color shift from yellow to green. Concurrently, ratiometric spectral changes at 380 and 725 nm for UV-Vis and signal enhancement at 755 nm for fluorescence were observed.
