[Determination of cinobufagin and resibufogenin in liver tissue by HPLC-MS/MS].

Objective: To develop a sensitive and accurate assay for detecting cinobufagin and resibufogenin in liver tissue using high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS).

Methods: The homogenization of liver tissue with internal standard dexamethasone was extracted with dichloromethane. The extracts with methanol were purified through ProElut C18 solid phase extraction and tested in positive electrospray ionization with multiple reaction monitoring of HPLC-MS/MS.

Results: The good linear relationship of cinobufagin and resibufogenin in liver tissue were 1-204 ng/g and 1-206 ng/g, respectively. The minimal detection threshold (S/N > or = 3) of this method was 0.3 ng/g for both cinobufagin and resibufogenin. The matrix effect was 96.5%-126.7%. The extraction recovery coefficient was 70.0%-82.3%. The precision of intra-day and inter-day was less than 10%.

Conclusion: This method is sensitive and reliable, and can be used in forensic toxicological analysis.

Xueshuan Xinmaining Capsules were qualitatively analyzed by UPLC-Q/TOF-MS for the first time, and 61 compounds were identified, including 9 phenolic acids, 10 bufadienolides, 15 saponins, 5 bile acids, and 22 other compounds. The chemical composition was slightly different between Xueshuan Xinmaining Tablets and Capsules.

Toad Venom (TV) is the dried product of toxic secretions from Cantor (BgC) or Schneider (BmS). Given the increasing medical demand and the severe depletion of wild toads, a number of counterfeit TVs appeared on the market, posing challenges to its quality control. Bufonis Venenum, an animal medicinal material, is widely used for treating cardiovascular diseases and pain induced by rheumatics or malignant tumors. In view of the high activity and high toxicity, it is of great significance to pay attention to the quality control of Bufonis Venenum to ensure the safety and effectiveness of its preparations. China's drug standards involve 102 preparations(474 batch numbers) containing Bufonis Venenum approved for sale, including 14 preparations in the Chinese Pharmacopoeia(2020 edition) and 68 preparations in the standards issued by the Ministry of Health Drug Standard of the People's Republic of China.
