Allergic patients are characterized by complex and patient-specific IgE sensitization profiles to various allergens, which are accompanied by different phenotypes of allergic disease. Molecular allergy (MA) diagnosis establishes the patient's IgE reactivity profile at a molecular allergen level and has moved allergology into the "Precision Medicine" era. Molecular allergology started in the late 1980s with the isolation of the first allergen-encoding DNA sequences.

(1) Background: In drug discovery and pharmaceutical quality control, a challenge is to assess protein extracts used for allergy therapy and in vivo diagnosis, such as prick tests. Indeed, there are significant differences between the features of marketed products due to variations in raw materials, purification processes, and formulation techniques. (2) Methods: A protein array technology has been developed to provide comprehensive information on protein-biomarker interactions on a large scale to support the pharmaceutical industry and clinical research.

The development of rapid analysis of human serum for the presence of allergen-specific Immunoglobulin E (IgE) is currently important. Consequently, we developed two types of three-dimensional (3D) protein biochips. The first one is a 3D hydrogel biochip containing hydrogel droplets with protein molecules (allergens, immunoglobulins and others).

Background: The role of autoimmune IgE responses in atopic dermatitis (AD) is highly debated. While IgE targeting self-proteins has been extensively studied, IgE responses induced by human-homologous exogenous molecular allergens (HEMAs) remains less understood.

Aim: To investigate whether IgE antibody responses to HEMAs are associated with AD, its severity, and response to dupilumab.
