(99m)Tc tricarbonyl glycine monomers, trimers, and pentamers were synthesized and evaluated for their radiolabeling and in vivo distribution characteristics. We synthesized a (99m)Tc-tricarbonyl precursor with a low oxidation state (I). (99m)Tc(CO)3(H2O)3 (+) was then made to react with monomeric and oligomeric glycine for the development of bifunctional chelating sequences for biomolecules. Labeling yields of (99m)Tc-tricarbonyl glycine monomers and oligomers were checked by high-performance liquid chromatography. The labeling yields of (99m)Tc-tricarbonyl glycine and glycine oligomers were more than 95%. We evaluated the characteristics of (99m)Tc-tricarbonyl glycine oligomers by carrying out a lipophilicity test and an imaging study. The octanol-water partition coefficient of (99m)Tc tricarbonyl glycine oligomers indicated hydrophilic properties. Single-photon emission computed tomography imaging of (99m)Tc-tricarbonyl glycine oligomers showed rapid renal excretion through the kidneys with a low uptake in the liver, especially of (99m)Tc tricarbonyl triglycine. Furthermore, we verified that the addition of triglycine to prototype biomolecules (AGRGDS and RRPYIL) results in the improvement of radiolabeling yield. From these results, we conclude that triglycine has good characteristics for use as a bifunctional chelating sequence for a (99m)Tc-tricarbonyl- based biomolecular imaging probe.

We report the use of unprotected amino acids as submonomer reagents in the solid-phase synthesis of -substituted glycine peptoid oligomers. Subsequent coupling of an amine, alcohol, or thiol to the free carboxylate of the incorporated amino acid provides access to peptoids bearing amides, esters, and thioesters as side chain pendant groups and permits further elongation of the peptoid backbone. The palette of readily obtained building blocks suitable for solid-phase peptoid synthesis is substantially expanded through this protocol, further enhancing the chemical diversity and potential applications of sequence-specific peptoid oligomers.

The intricate relationship between brain vascular diseases and neurodegeneration has garnered increased attention in the scientific community. With an aging population, the incidence of these two conditions is likely to increase, making it imperative to understand the underlying common molecular mechanisms and unveiling novel avenues for therapy. Prompted by the observation that Aβ peptide aggregation has been implicated in the development of cerebral amyloid angiopathy (CAA) and that elevated concentrations of vascular endothelial growth factor (VEGF) in the cerebrospinal fluid (CSF) have been correlated with less cognitive decline in Alzheimer's disease (AD), we demonstrate that a small peptide (Pep9) encompassing the 10-30 sequence of VEGF exhibits significant ability to inhibit the aggregation of the Aβ1-42 peptide, as well as the formation of toxic oligomers.

Understanding how a macromolecule's primary sequence governs its conformational landscape is crucial for elucidating its function, yet these design principles are still emerging for macromolecules with intrinsic disorder. Herein, we introduce a high-throughput workflow that implements a practical colorimetric conformational assay, introduces a semi-automated sequencing protocol using MALDI-MS/MS, and develops a generalizable sequence-structure algorithm. Using a model system of 20mer peptidomimetics containing polar glycine and hydrophobic -butylglycine residues, we identified nine classifications of conformational disorder and isolated 122 unique sequences across varied compositions and conformations.
