Objective: To analyze the transcription factor binding sites and methylation in the promoter region of UCA1 gene.
Methods: UCA1 gene promoter was analyzed using CpG software to predict the CpG island. MatrixCatch and TFSEARCH were used to predict the potential transcription factor binding sites in UCA1 gene core promoter. ChIP assay was used to identify the transcription factors binding to UCA1 gene core promoter.
Results: UCA1 gene promoter contained no CpG island and was therefore a typical tissue-specific gene. There were 4 transcription factors associated with human cancers in UCA1 gene core promoter, but only one of them interacted with UCA1 gene core promoter.
Conclusion: There is no CpG island in UCA1 gene promoter region, and the transcription factor c-Myb can specifically bind to the core promoter.

Metabolic reprogramming is a hallmark of cancer, and abnormal lipid metabolism is associated with drug resistance in bladder cancer cells. The long noncoding RNA (lncRNA) UCA1 is overexpressed in bladder cancer, but its functional contribution to lipid metabolism remains uncharacterized. In this study, we demonstrated that lncRNA UCA1 inhibits epirubicin-induced cell apoptosis by supporting abnormal lipid metabolism in bladder cancer cells.

Programmed cell death (PCD) is a process that eliminates infected, damaged, or possibly neoplastic cells to sustain homeostatic multicellular organisms. Although long noncoding RNAs (lncRNAs) are involved in various types of PCD and regulate tumor growth, invasion, and migration, the role of PCD-related lncRNAs in bladder cancer still lacks systematic exploration. In this research, we integrated multiple types of PCD as pan-PCD and identified eight pan-PCD-related lncRNAs (LINC00174, HCP5, HCG27, UCA1, SNHG15, GHRLOS, CYB561D2, and AGAP11).
