Article Abstract

A total of eight cellular alterations associated with human carcinogenesis have been framed as the 'hallmarks of cancer'. This representation overlooks a ninth hallmark of cancer: the requirement for tumor-originating distributed stem cells to shift sufficiently from asymmetric to symmetric self-renewal kinetics for attainment of the high cell production rate necessary to form clinically significant tumors within a human lifespan. Overlooking this ninth hallmark costs opportunities for discovery of more selective molecular targets for development of improved cancer therapeutics and missing cancer stem cell biomarkers of greater specificity. Here, the biological basis for the ninth hallmark of cancer is considered toward highlighting its importance in human carcinogenesis and, as such, its potential for revealing unique molecules for targeting cancer diagnostics and therapeutics.

Background: Mitochondrial dysfunction has been demonstrated to be an important hallmark of sarcopenia, yet its specific mechanism remains obscure. In this study, mitochondrial-related genes were used as instrumental variables to proxy for mitochondrial dysfunction, and summary data for sarcopenia-related traits were used as outcomes to examine their genetic association.

Methods: A total of 1,136 mitochondrial-related genes from the human MitoCarta3.

Article Synopsis
  • Histone lysine demethylase LSD1 (KDM1A) is identified as a key regulator in cancer, particularly for enhancing T cell activity and anti-tumor immunity, though its effects on the tumor microenvironment in esophageal squamous cell carcinoma (ESCC) are not well understood.
  • The study utilized various methods, including tissue microarray immunostaining, colony formation assays, and advanced genomic techniques, to analyze KDM1A's role in ESCC and found it correlates with patient survival, inhibiting cancer cell growth when blocked.
  • KDM1A removes specific methyl markers from histones, thereby reactivating transcription of the RAD51 gene, while also inhibiting proinflammatory gene expression

Remodelling of T-Tubules and Associated Calcium Handling Dysfunction in Heart Failure: Mechanisms and Therapeutic Insights.

Electronic address:

In cardiomyocytes, transverse tubules (T-tubules) are sarcolemmal invaginations that facilitate excitation-contraction coupling and diastolic function. The clinical significance of T-tubules has become evident in that their remodelling is recognised as a hallmark feature of heart failure (HF) and a key contributor to disrupted Ca homeostasis, compromised cardiac function, and arrhythmogenesis. Further investigations have revealed that T-tubule remodelling is particularly pronounced in HF with reduced ejection fraction (HFrEF), but not in HF with preserved ejection fraction, implying that T-tubule remodelling may play a crucial pathophysiologic role in HFrEF.

  • Cellular senescence is a dual-edged sword in cancer, acting as a protective barrier before cancer develops but potentially promoting tumor growth once cancer is present.
  • The paper highlights how the senescence-associated secretory phenotype contributes to the diversity of senescent cells in tumors and discusses various triggers for cellular senescence, along with emerging senolytic drugs aimed at clearing these cells.
  • Overall, the research sheds light on the complex roles of senescent cells in different cancers and offers new perspectives for developing cancer therapies.

Electronic address:

Fibrosis refers to the progressive tissue lesion process characterized by excessive secretion and deposition of extracellular matrix (ECM). Abnormal fibrous tissue deposition distorts tissue architecture and leads to the progressive loss of organ function. Notably, fibrosis is one of the primary pathological appearances of many end stage illnesses, and is considered as a lethal threat to human health, especially in the elderly with ageing-related diseases.
